Objective: To study the effect of tanshinone Ⅱ-A(Ts Ⅱ-A)on proliferation and differentiation of rat osteoblast in vitro and expression of BMP-2 and TGF-β1 m RNA, and explore the possible mechanism, which may provide experimental basis for Ts Ⅱ-A on orthodontic tooth movement. Methods: Rat osteoblast was isolated, cultured and purified in vitro. And we identified the morphology of third passage of osteoblast. Then osteoblast was divided into experimental group and control one. The former was treated with Ts Ⅱ-A at 5×10-3 mg/L, 5×10-2 mg/L, and 5×10-1 mg/L respectively, while the latter was cultured with DMEM. The proliferation of osteoblast was tested and expression of BMP-2 and TGF-β1 were detected by RT-q PCR at day 1, 3, 5 and 7 separately. ALP was also examined at day 5. Results: The inverted microscope observation and ALP and Alizarin Red staining showed that the morphology and biological characteristics of the cultured cells were in accordance with these of osteoblast. Cell Counting Kit-8(CCK-8)detection showed that OD values of each experimental group were greater than those in control group, especially the experimental group at concentration of 5×10-2mg/L. And OD values of ALP in experimental group were also greater than these in control group(P<0.05). In experimental group the expressions of BMP-2 and TGF-β1m RNA were increased when compared to the expressions in control group(P<0.05). Simultaneously, the expression in experimental group increased as time passed, and the differences were statistically significant(P<0.05). Conclusions: Ts Ⅱ-A could promote the proliferation and differentiation of osteoblasts within a period of time and also could up-regulate the expression of BMP-2 and TGF-β1 m RNA, which may make Ts Ⅱ-A play a role in the orthodontic tooth movement during bone reconstruction. |
