Objective:1,Through comparing the observation of the appearances,histopathological features,the healing percentage changes,as well as the changes of TXB2 and MVD levels in the early wound of deep second degree burn rats,to learn the patterns of the microcirculation changes in the burn wound at early period.2,To observe and study the variation of the appearances,histopathological features,the healing percentage changes,as well as the levels of TXB2 and MVD change in the early wound tissue of deep second degree burn rats after using PGE1 in early stage.And explore the influence of wound microcirculation while using PGE1 in early stage of deep second degree burn rats wound.Thus,we are able to provide a new thought which can improve the prognosis of deep second degree burn wound,promote wound healing and enhance the clinical curative effect in burn wound.Materials And Methods:90 SPF level of SD rats(wighted 200+20)from the Hunan SJA Laboratory Animal Co.,Ltd were includes in theexperimental,with a male-female ratio of50—50.Randomly divided this rats into 3 groups: false burn group(n = 30),burn control group(n = 30),burn with PGE1 treament group(n = 30).The rats in the experiment for a acclimated raise after a week,then give each group respectively sodium sulfide to depilatory and chloral hydrate to anesthesia.Give the burn control group and the burn with PGE1 treament group 5 seconds steam with 4 Mpa pressure,106 ℃,leading to a 30% TBSA II ° scald in rats.The rats accepted the fluid infusion therapy after the procedure.The false scald group rats were subjected with37 ℃ water mist spraying for 5 seconds.Then we gave to the wound of the rats iodophor,BID,for disinfection.The false scald group and the burn control group were both giving intraperitoneal injection of 1 ml of 0.9% saline,the PGE1 treatmentgroup was giving an intraperitoneal injection of 1ml of PGE1(1.6μg/kg)mixd with saline simultaneously.Until rats were executed,the indentical injection of each group were repeated at 8 am everyday.Observed and recorded general wound apperances of each group,then randomly selceted 10 rats in each group to calculate the percentage of wound healing on 3rd,7th 14 th day.We draw 5ml abdominal aortic blood after anesthesia in aseptic condition,and collected the blood samples in the aseptic procoagulant tube at room temperature for 30 min,then collected the serous after centrifuged with 4000 r/min for 5 minutes,used the ELISA method to detect the TXB2 of the serous samples and rapidly perfused normal saline to the heart of rats at the same time,until the lung tissue get pale,then cut off the dorsal wound tissue of rats,soaked them in formaldehyde and used immunohistochemical method to detect the expression of CD31 to calculated the density of microvascular,and employed HE method to observe the pathological changes of the wound in every period.Results:1,General observation of wound in Each group: The dorsal skins in false scald group were normal,without obvious changs.After 3 days,black spots were found in the wound of burn with PGE1 treament group wound,without exudation.The most part of burn control group’s wounds change to black,and part of them formed to eschar,with some exudations.On the 7th days,part of wounds in the PGE1 treament group became to be dry scab,the wound extent was reduced than before,then we could see a little effusions in burn control group wound,which became black,and eschar was slightly shrink than before.On the 14 th day;the PGE1 treament group wound was obviously shrink and dry,without exudation;the wound of burn control group was still black,there was slightly contracted eschar with big black necrotic defect became into integrated,and a small amount of purulent secretion around the necrotic wound.2,Comparison of pathologic morphology in each group wound tissue : There were no distinct abnormity about pathologic morphology in false scald group.On the3 rd day,at the burn with PGE1 treatment group,we could see that new epithelium began to grow,and there were a lot of necrotic tissue and some infiltratedinflammatory cells;On the 7th day,the epithelial tissue become obviously thicken in burn with PGE1 treatment group.there was neogenesis of epithelial tissue in the burn group,and the epithelial tissue was thin and unapparent;On the 14 th day,the new epithelium became mature,and most part of the wound were healed in the burn with PGE1 treatment group.In the burn control group,the new epithelial tissue were still thin,and there was no formation of epidermis tissue.3,Wound healing rate in each group: on the 3ird day,there was no difference between the burn with PGE1 treatment group and the burn control group about the wound healing rate;We could find out that wound healing rate of PGE1 treatment group were significantly higher than the burn control group(P<0.05),The wound healing rate had no obvious change in false burn group.4,Comparison of serous TXB2 in each group:(1)The serous TXB2 showed a decreasing trend over time in burn with PGE1 treatment group,while there was a rising trend in burn control group.(2)Compared with the false burn group,the level of serous TXB2 was higher at every time phrase in burn with PGE1 treatment group.(P<0.05).on the 14 th day,the level of TXB2 in the burn with PGE1 treatment was close to the false burn group,and the level of TXB2 was still significantly higher in burn control group than which in false burn group.(3)Compared with the burn control group:the level of serous TXB2 in the burn with PGE1 treatment group was obviously lower than control group(P<0.05)on the3 rd,7th,14 th day.5,The microvessel density(MVD)in every group: There was no neovascularization in the false burn group.The expression of the microvessel density(MVD)was manifested an uptrend over time in all group.The MVD in PGE1 treatment group was significantly higher than the burn control group(P<0.05)on the3 rd,7th,14 th day.Conclusions:1,There was an obvious dysfunction that gradually became exacerbated in microcirculation of deep second degree burn wound of rats which could be the mainreason cause the deterioration in early stage of burn wound.2,The early intervention of PGE1,which is able to significantly ameliorate burn wound microcirculation,promote microvascular generation,reduce platelet aggregation,improve wound prognosis,accelerate wound healing,can be popularized and applied in clinic.
