Corydalis yanhusuo is a representative drug of vinegar preparation method of Chinese medicine processing.Vinegar is processed into the liver meridian is one of the important contents of the processing theory.In this paper,We studied the effect of the Main Active Ingredient Content,Pharmacodynamics and metabolic regulation of main componentsthe contents of the chemical components,the changes of pharmacodynamics indexes and the main components of the metabolic law of Corydalis yanhusuo before and after vinegar processing on the basis of Corydalis yanhusuo processing technology specification,to investigate the Corydalis treated with vinegar,the liver meridian effect.First of all,We selected the best processing technology of vinegar Rhizoma corydalis yanhusuo by means of orthogonal test method.The factors are the amount of auxiliary materials,the temperature of stir frying,the time of frying and the frequency of stir frying.Index is the Main Ingredient Content of Tetrahydropalmatine(THP),D-corydaline(DHC)、 Protopine(PRO)and so on.Results show that each 100 grams of Corydalis yanhusuo with 20 ml vinegar,sealed infiltration 30 minutes,150-160 ℃ temperature and at a frequency of 40 times per minute turn fried 6min is the best one.The main active components of the samples were determined by the method of high performance liquid chromatography.Tetrahydropalmatine growth of30.09%,D-corydaline growth of 23.56%,Coptisine growth of 20.02% and total alkaloids growth of 19.85% are the results of corydalis yanhusuo processed by vinegar,which is verified that the vinegar increases the effective components dissolution of Corydalis and plays a synergistic role.Second,the acute hypostasis model of rats were subcutaneously injected adrenaline to simul state of rage and soak of iced water to simul state of cold-evil,and thehemorheologic parameters were detected.Results show that different doses of Corydalis crude and vinegars products on blood rheology of blood stasis model rats index improved in varying degrees.The indicators of vinegars products of the same dose improved refluxing effect is stronger than crude product.Vinegar product of the same dose have more significant difference than raw product group,and the role of vinegars products in removing blood stasis is stronger than that of crude product from the indicators of Whole blood viscosity(200s-1 、 50s-1),Blood sedimentation equation K value,erythrocyte aggregation index and erythrocyte rigidity index.It’s demonstrated that vinegar-processed Corydalis yanhusuo group improved in varying degrees of hemorheological indicators of blood stasis rats and enhanced the effect of activating blood circulation to remove blood stasis.Many activating blood circulation and removing blood stasis drugs are into liver meridian,have effects of soothe liver,regulate Qi and eliminating stasis to stop pain.The experiment verified that Corydalis yanhusuo is vinegar-processed into the liver meridian and enhancing efficacy.Third,It’s found that crude corydalis yanhusuo and vinegar products could reduce the index of liver coefficient,liver microsomal protein content and CYP450 enzyme content through to the main impact factor for testing in the liver.In particular,the inhibitory effect of vinegar products on the content of CYP450 enzyme content was obvious.Results show that the high dose of crude corydalis yanhusuo and vinegar products have a significant reductionin the index of liver coefficient,liver microsomal protein content and CYP450 enzyme content of rats compared with that of health control.Especially the CYP450 enzyme content has a significant inhibitory effect,in which the effect of vinegar is more significant.It’s a comprehensive analysis that drug groups could reduce the detection index,especially liver microsomal protein content and CYP450 enzyme content decreased,indicating that the activity decreased,suggesting that of Rhizoma Corydalis on liver drug metabolizing enzyme is enzyme inhibition of the trend.It’s speculated that corydalis yanhusuo of synergism might be related to the inhibition of hepatic drug metabolizing enzyme activity,medication slows metabolism,thus making it prolonged retention time related.Finally,the tetrahydropalmatine single intragastric administration in rats at different time points were detected the content of tetrahydropalmatine in blood through the pharmacokinetic method and HPLC method,then Blood drug concentration-time profiles were obtained.The results showed that tetrahydropalmatine reached a maximum concentration of 0.219 g·ml-1 at about 0.75 h.After that,its content began to fall and some unknown peaks appeared.We got the analysis of Substance change process of tetrahydropalmatine in vivo Through liquid mass spectrometry.The results showed that the main component is the prototype of tetrahydropalmatine into the blood.After a certain period of time,A part of tetrahydropalmatine decomposed.Presumably,two possible products were produced.Their retention time is 2.28min(A)、17.31min(B).These two substances have more than half of the fragment ion peaks can be consistent with tetrahydropalmatine through preliminary analysis of spectral analysis.It is believed that These two substances may be the metabolic product of tetrahydropalmatine.By extrapolating the atlas,There are two possible structures of product A that 11,12 were replaced by methylene-dioxy(-O-CH2-O-),6 or 8 were replaced bymethoxy(-OCH3)in the tetrahydropalmatine structure.There are Twelve possible structures of product B.Two positions were replaced by carboxyl(-COOH)and carboxymethyl(-OOCHCH3)at four possible substitution positions in C(5,6,8,13C).This result provides the possibility of the material basis for the study of Tetrahydropalmatine in vivo transformation and Corydalis vinegar-processed into the liver.Briefly,in this paper,it is verified and explained for traditional Chinese medicine Rhizoma Corydalis vinegar processed into the liver by the mechanism,using modern science and technology through the above four parts.It is a systematic interpretation of the theory of traditional Chinese medicine as a case.
