Significance Of Klf6 Expressed In Human Colonic Carcinoma And Precancerous Lesions

Objective:To investigate the expression of kruppel-like factor 6 in tissues of the colon cancer,precancerous lesions and in normal cells.To study the kruppel-like factor 6expression level of mutual relations in all types of organizations,and the function in colon cancer progression.Methods: To collect 88 cases of colorectal cancer samples from First Affiliated Hospital of Wannan Medical College,including pathological confirmed for 46 cases of cancer tissue and 21 cases of precancerous tissue.And take away from the edge of cancer more than 5 cm of normal colorectal tissues adjacent to carcinoma as the control group.After in vitro were divided into two parts,one part from the body about 500 mg put into-80 ℃ refrigerator within 1 hour.Q-polymerase chain reaction(PCR)to detect KLF6 expression level in colorectal cancer(colorectal cancer group),Precancerous lesions(Precancerous lesions group)and Tissue adjacent to carcinoma(control group).The Most specimens were sent to the General Department of Pathology,After taking the pathological paraffin blocks,then by using immunohistochemical SP method research the expressions of KLF6 protein in Colorectal cancer,precancerous lesions and normal human cells.Results: 1.immunohistochemical results:1.1KLF6 protein expression in the nucleus,cytoplasm and cell membrane of colorectal cancer cells,precancerous cells and normal colorectal mucosa.KLF6 expression in colorectal cancer tissue level is significantly lower than that of normal tissue adjacent to carcinoma(P < 0.05);KLF6 expression level in colorectal cancer precancerous tissue also significantly lower than the normal tissue adjacent to carcinoma(P < 0.05);It is no significant difference between colorectal cancer tissue and precancerous tissue of expression of KLF6(P>0.05).1.2 KLF6 expression with Tumor infiltrating,extent of differentiation,lymph node metastasis and TNM stages have certain relevance(P<0.05);KLF6 expression with Patient’s age,sex,tumor histological type,and the size of the cancer has no obvious correlation(P>0.05).2.The results of Q-PCR:The gene expression was described by relative expression levels.The results showed that KLF6 gene has a lower level of expression in colorectal cancer than that in normal tissues.The difference was statistically significant(P<0.05);KLF6 genes expression in normal tissue is higher than the precancerous tissue,the difference was statistically significant(P < 0.05);KLF6 gene expression quantity in precancerous tissue higher than that of colorectal cancer tissues,the difference was statistically significant(P < 0.05).Conclusion: KLF6 expression in colorectal cancer tissue level is significantly lower than that of normal tissue adjacent to carcinoma(P < 0.05);KLF6 expression level in colorectal cancer precancerous tissue significantly lower than the normal tissue adjacent to carcinoma(P < 0.05);The expression of KLF6 in colorectal cancer tissue and precancerous tissue need further confirmed.KLF6 expression with Tumor infiltrating,extent of differentiation,lymph node metastasis and TNM stages have certain relevance(P<0.05).
