Ershen pill comes from “Pujibenshi prescription” written by Xu Shuwei in the Song Dynasty,which consists of fructus psoraleae and nutmeg. It is the basic treatment of insufficiency of the spleen and diarrhea in clinic due to its function of warming the kidney and spleen and relieving diarrhea. Diarrhea of which weak-spleen diarrhea is in primary proportion is a familiar chronic spleen and gasoric disease. Sishen pill is extensively applied in treating weak spleen and kidney diarrhea,also called five night watches-diarrhea.Curative effect is obvious. But Comparative study on effects of the alcohol extracts from Psoralea corylifolia L﹠Nutmeg to treat spleen deficiency diarrhea. The influence of different dose of ershen pill on the morphological changes of gastrointestinal tract function of Intestinal propulsion, immunologic function, AQP, Gastrointestinal function of the model mouse with insufficiency of the spleen and diarrhea is studied in this issue to research ershen pill from several aspects to provide experimental basis for clinical medication.Objective:To study the effect of psoralen nutmeg on spleen deficiency models and immunological functions in mice Methods:Kunming mice were randomly divided into six groups, including normal group, model group,alcohol extracts from Psoralea corylifoliaL ﹠ Nutmeg(21.06 g/kg, 14.04 g/kg, 7.02 g/kg), and ShenLingBaiZhu(2.34g/kg). Except for nomal group, the rest groups were given water decoction of rheum officinale-induced spleen deficiency diarrhea in mice. It has been used to diarrhea of rat model suffered from spleen deficiency after taking Rhubard; The application of charcoal powder propelling method for the determination of intestinal propulsive; Optical microscope observation of stomach intestinal tissue morphological changes; The content of D-xylose, amylase, NO, NOS,MDA and SOD were checked by biochemical method; The enzyme- linked immunosorbent assay(ELISA) was used to detect the contents of SP, IL-2, TNF-a, VIP and IL-1 in the colon of mice; The protein expression level of AQP3 and AQP4 were measured by immunohistochemistry; real-time fluorescent quantitative PCR was performed to detect the expressions of AQP3, AQP4 mRNA,andWestern blotting to determine the expressions of AQP3, AQP4 protein.Results:It is showed by optical microscope that the alcohol extracts from Psoralea corylifolia L ﹠Nutmeg have different degrees of improvement to intestinal injury.meanwhile, Compared with model group, the alcohol extracts from Psoralea corylifolia L ﹠ Nutmeg not only effectively increased levels of NO, NOS, SOD, D-xylose, amylase, elevated expressions of AQP3 and AQP4,but also inhibited expressions of MDA, IL-1β, IL-2, TNF-a, SP and VIP in the mice.Conclusion:The alcohol extracts from Psoralea corylifolia L﹠Nutmeg has certain treatment function on model of insufficiency of the spleen and diarrhea. It can not only effectively improve the overall situation of mice, but also significantly enhance the intestinal antioxidant capacity and reduce the lipid peroxidation damage, which might be related to regulate the expressions of NO, NOS, SOD,IL-1β, IL-2, TNF-a, SP, VIP, AQP3 and AQP4 in the mice.
