Study On The Anti-liver Cancer Activity Of Traditional Chinese Medicine LH Prescription

Liver cancer is one of the main malignant tumors, and more than half of global new cases in our country. In contrast, the chemical drug is scarce for treatment of liver cancer besides Sorfenib. As more attention on survival with tumor and the qulity of life, Traditional Chinese medicine(TCM) has played a more and more important role.LH compound, composed of Paris Poiyphylla and Carcuma Longa, is the preious research results of our research group, which has the inhibitory rate of 50%. In this research, the tumor-bearing mice models of H22 hepatocarcinoma and Lewis lung adenocarcinoma in mice-derived, Hep G2 hepatocarcinoma and NCI-H460 lung adenocarcinoma in human-derived were utilized to appraise the anti-tumor activity of LH compound. Meanwhile, in order to evaluate the life quality of tumor-bearing mice, the water and food intake and the degree of fatigue were calculated. The immune function was reflected by viscera index, such as spleen index, thymus index etc.Curcumin is the main active ingredient in turmeric. The Everted Sac Technique results showed that the 0.4 mM of curcumin combined with PRS can significantly increase the absorption of RPS in the duodenum and the total absorption in the small intestine, and raised the apparent permeability coefficient of RPS.Although LH showed great tumor suppression effect, the side-effect of toxicity and reducing the body weight of mouse should be alleviated though combined with other traditional Chinese medicine. After systematic screening of traditional Chinese herb, Rhizoma Anemarrhenae has been found to combine with LH, which called LHZM, not only overcame the LH limitations but also improved its anticancer effect. Therefore, it’s significant to investigate the anti-tumor effect of the compound of LHZM. The research suggested that LHZM compound showed more outstanding anti-tumor effect. The inhibitory rate on H22 and EAC were improved from 56.86%, 47.95% to 63.25%, 58.99% respectively. In addition, LHZM showed good analgesia and hypnotic effect though scramble net experiment, analgesic experiment and the sleep experiment of mice induced by sodium pentobarbital, which might be connected with the Rhizoma Paridis saponins(RPS) and timosaponin in LHZM. And compared with the LH compound, the body weight of mouse was increased significantly after oral administration LHZM. In conclusion, the present of our research provided theoretical and practical basis for LHZM application.Single index(inhibitory rate or prolong survival time) cannot fully reflect the superior effect of drugs. The comprehensive evaluation, considering inhibitory rate, prolong survival and the symptoms such as pain, fatigue, immunity, appetite and sleep quality index. Therefore, this article hopes that the counterparts in the study of anti-tumor drugs can pay more attention on the comprehensive index evaluation.
