Dioscorea persimilis(DP) belongs to Dioscorea genus, Sect. Enantiophyuum Uline, which often used as Dioscorea opposita(DO) at the local level. The chemical constituents of DP have been reported rarely, moreover, t here are serious different opinions on the quality evaluation of DP and the rationality of DP used as DO. Therefore, we conducted studies on chemical constituents of DP and its potential pharmacological activities, and compared chemical constituents and pharmacological activities of DP with DO.Chemical constituents of the ethanol extract derived from DP were isolated and purified through conventional methods. 12 compounds were isolated and 9 of them have been identified as β-sitosterol, stigmasterol, β-sitosteryl palmitate, methyl heptadecanoate, glyceroyl monopalmitate, β-sitosteryl-3β-glucopyranoside-6’-Opalmitate, vanillic acid, syringic acid, cyclo(L-Phe-L-Tyr). Each fraction of the extract derived from DP was presented significant anti- inflammatory and immune-reglation activities, and the main components of fractions were extreme variation. Furthermore, the potential correlation between pharmacological activities and main chemical composition was analysed. Anti- inflammatory acivity was closely related to the total triterpenes(r = 0.988) in egg albumin induced paw swelling experiment and total flavonoids(r = 0.996) in xylene- induced ear swelling experiment, while inmmune regulation was highly correlation with total saponins(r = 0.907).In addition, the content of total triterpenes, total saponins and total phenolic acids in DO were also significantly higher than DP. The main active compounds might be gallic acid, chlorogenic acid, saponin through UPLC-Q-TOF analysed, the different of active components content lead to different activity. Pharmacological activities(including antioxidant, anti- inflammatory and immune regulation) of DO were better than DP at the same dosage. There are differences between DP and DO cannot be ignored. So DP instead of DO as herb medicine is obviously insufficient.
