Objective:To observe low strength of light to the mouse retina,then further study of the pathogenesis and intervening measures of visible light to retina can be done.Methods:There are 30 C57BL/6J mices, same week old wild type,recorded their weight,divided into two groups randomly.The experiment group is in dark adaption at 5:00-6:00 every day.,then they were lighted at 6:00-7:00 pm by 100 lux LED.While the control group is only in dark adaption at particular time everyday.At 1,3,7,14,30 days after the beginning, the mouses were anesthetized with intraperitoneal injection of a mixture of ketamine and xylazine(equal mixture of ketamine and xylazine,0.15ml/kg),then flash electroretinogram(fERG)is done on these mouses.Afterwards kill them via cervical dislocation,then their eyes were enuncleated and processed to frozen section.HE coloration is done on these eyeballs.Every layer of the construction of the ratina and the thickness of the outer nuclear layer(ONL) could be determined.Use SPSS for data statistics, with the results in the form of X ±S. Groups were analyzed using unpaired t test. P<0.05 was considered statistically significant.Results:Through the observation,compared with the control group,the first day of the lighting,the implicit time of Rod-R of a wave in test group were 23.2+ 2.726ms,the control group were 19.3±1.546ms,the implicit time of Cone-R of b wave in test group were 33.9±4.658ms, the amplitude were 53.8±3.185uv, while the control group were 24.3±3.287ms,62.3±2.754uv;3 days after the beginning,the implicit time of Rod-R of a wave in test group were 20.9± 2.726ms,the control group were 18.3±1.546ms,,the implicit time of Cone-R of b wave in test group were 32.9±4.658ms, the amplitude were 52.8±3.185uv,while the control group were 27.3±3.287ms67.3±2.754uv;7 days after the beninning,the implicit time of Rod-R of a wave in test group were 20.9±2.726ms,the control group were 18.3±1.546ms, the implicit time of Cone-R of b wave in test group were 35.9±4.658ms,the amplitude were 48.8±3.185uv, while the control group were 24.3±3.287ms,62.3±2.754uv; 14 days after the beginning,the implicit time of Rod-R of a wave in test group were 25.8±2.726ms, the control group were 16.5 ±1.546ms,the implicit time of Cone-R of b wave in test group were 30.9± 4.658uv,the amplitude were 50.8±3.185uv,while the control group were 25.3± 3.287ms,66.3±2.754uv;30days after the beginning,the implicit time of Rod-R of a wave in test group were 23.9±2.726ms, the control group were 19.3±1.546ms, the implicit time of Cone-R of b wave in test group were 30.9±4.658ms, the amplitude were 50.8±3.185uv, while the control group were 25.3±3.287ms,66.3± 2.754uv.In the opinion of the five times,compared to the control group,the implicit time of Rod-R a wave in test group were longer,the implicit time of b wave of cone-R were longer,the amplitude were shorter.In terms of pathological histology,the layer of retina in experimental group were clear,cell nucleus of inner nuclear layer (INL)and outer nuclear layer(ONL) were intensive.The thickness of ONL were 31.3 ±2.3um,30.2±2.5um,30.6±1.6um,31.6±1.7um,30.5±1.3um.Conclusions:100 lux light could do damage to the retina of dark-adapted mouse.
