| ObjectiveStudy the effects of Vaccinium uliginosum(VU) to the electroretinogram(ERG) andretinal hisoopathological changes of rabbits before and after light-induced damage.Methods1. Animal model establishing:Based on the literatures, light-induced damage model was created by being exposed in a light box.2. Grouping:Thirty-five Chinchilla Rabbits were divided into five groups according to the randomization tables:administrated beforehand (A,B), light injury without administration (C), administrated after injury (D) and blank control (E).3. Examination of retinal function and histology:After administrating group A and B with Vaccinium uliginosum (VU) homogenate once a day for four weeks we observed the tissue & structure of the rabbits in group B and E under the light microscope and measured the thickness of their out nuclear layers (ONL) and apoptosis index (AI); all others'electroretinogram (ERG) were recorded according to the standards set by the International Society for Clinical Electrophysiology of Vision(ISCEV). Then group A, C and D were exposed to strong light. Also we stopped feeding group A with VU and started to feed group D with it. We recorded ERG of group A, C and D after 1 day,1 week and 2 weeks respectively. Finally we observed the tissues &structures of them and measured ONL and AI of them.4. SPSS 12.0 software package was used for one-way or double ways ANOVA and LSD test.Results1. After four weeks feeding with VU, the latency time of group A got shorter than group C and D, and the amplitude got increased. After exposing to strong light latency time lengthened and amplitude decreased in group A, C and D, but compared at each same moment the measured values of group A were better than group C. With the accumulation of VU the ERG of group D was improving and all of the detection values were better than group C at last.2. Group B and E were normal in histology and lagyers were in order. While group C got disordered. Group A and D were injured slightly. The thickness of ONL among all groups had statistical significance(F=330.506 P<0.01).The thickness of group C was(18.048±1.767)μm and thinner than others'(P=0.000).3. There was statistical significance among all groups in AI(F=230.126, P<0.01). AI of group C was (10.960±1.534)% and was higher than others'(P= 0.000).AI of group B was (1.817±0.203)% and lower than group E (P=0.000).Conclusions1. VU can reduce the apoptosis and relieve the harm of the rabbits' retina exposed to normal day and night.2. VU can relieve the harm of light when used beforehand.3. VU can repair the light damage to retina. |
