Study On Chemical Composition And Quality Control Of Qidengmingmu Jiaonang

As one of the complications of diabetes, diabetic retinopathy (DR) can make vision gradually decline and even blindness [1-3]. Qidengmingmu Jiaonang consists of effective parts from puerariae. astragali radix and erigerontis herba, Pharmacological studies have shown that [3,5], Qidengmingmu Jiaonang has exact curative effect on rats with diabetic retinopathy. But it hasn’t to develop its material basis research, also there is no relevant quality standards literature reports of it. In this paper, we have preliminarily studied its material basis and determinated its principal components, also we have found the composition of its effectiveness through the serum drug chemistry and establish its quality standard.Objective:1. To research the chemical composition of Qidengmingmu Jiaonang.2. To analysis the transitional elements in the blood of Qidengmingmu Jiaonang.3. To establish the quality standards of Qidengmingmu Jiaonang.Methods:1. Using mass spectrum scanning method to determinate its chemical molecular weight; Thenwe determined the possible chemical composition by relevant literature and database data; Last, with the reference substance, using HPLC method, we identify and determine its main components2. Using serum medicinal chemistry method, we analysis them with disassemble the prescription explore the effective parts to find the compositions which can get into blood and where they come from.3. Using Astragaloside IV and puerarin as qualitative indicators to establish TLC identification method; We got puerarin and scutellarin as quantitative indexes to set up content of HPLC determination method.Results:1. We have found Qidengmingmu Jiaonang contain ingredients are as follows.The extracts from puerariae contain puerarin and daidzein, maybe include daidzin, puerarin-4’-O-glucoside, daidzein-4’,7-diglucoside, genistin, mirificin, 3’-methoxydaidzein,3’-methoxydaidzin, ononin, biochanin. The extracts from erigerontis herba contain scutellarin,3,5-O-dicaffeoylquinic acid (isochlorogenic A), luteolin-7-glucuronide,4,5-O-dicaffeoylquinic acid (isochlorogenic C). 3,4-O-dicaffeoylquinic acid (isochlorogenic B); maybe include 4-O-caffeoylquinic acid, chlorogenic acid. 1-O-methyl-3,5-O-dicaffeoylquinic acid methyl ester. 1,3-O-dicaffeoylquinic acid.2. Serum medicinal chemistry research shows that the compounds which can get into blood are puerarin, daidzein (which come from the extracts of puerariae) and scutellarin (which come from the extracts of erigerontis herba)3.Its TLC identification had a good resolution with clear spots; Puerarin, sctellarin showed a good linearity in the ranges of 0.005-0.040 mg·mL-1(r=0.9999),0.002-0.058 mg·mL-1 (r=0.9998). The average recoveries were 99.38%(RSD=1.12%, n-9),98.04%(RSD=1.44%, n=9).Conclusion:1. The content of sctellarin, isochlorogenic A, isochlorogenic B, isochlorogenic C, luteolin-7-glucuronide, daidzein in Qidengmingmu Jiaonang are respective in the ranges of 0.98%～1.04%,0.44%～0.52%,0.15%～0.21%,0.81%～0.98%, 1.15%～1.18%.2. Puerarin, sctellarin, daidzein which can get into blood after oral administration perhaps are Qidengmingmu Jiaonang’s therapeutic material basis.3. The established standard is acceptable for quality evaluation of Qidengmingmu Jiaonang.
