Objective:To observe the different response of local purines release between acupuncture and moxibustion.Methods:SD rats were selected to study the different response of local purines release between acupuncture and moxibustion.The SD rats in physiological condition were divided into control, acupuncture with rotating, acupuncture without rotating and control with moxibustion group, while the pathological condition including model, CFA with acupuncture-rotating, CFA with acupuncture-without-rotating and CFA with moxibustion group. Complete Freund’s adjuvant (CFA) was applied to induce inflammatory pain.Acupuncture or moxibuation treatment were carried out at rats’ local "Zusanli" point.The microdialysates were collected before, during and after the manipulation over a 30min period for a total of two and half hours. The high-performance liquid chromatography(HPLC) was utilized to analyze ATP and adenosine in the local dialysates. The change rate at each point compared with baseline were calculated for statistical analysis with SPSS 17.0 statistical software.Results:1. The changes of local purines under physiological condition1) The change rate of ATP level in acupuncture with rotating group increased 55% at 30min and decreased at 60min when compared with control group (P<0.05),while the change rate of ATP level in control with moxibustion group shows difference at 60min for 24% with a delayed elevation(P<0.05).There were no differences among other inter-groups at each time point (P> 0.05).2) The change rate of ADO level in acupuncture with rotating group rised 179% and 107% at 30min and 60min respectively(P<0.01) and acupuncture without rotating group showed increase at 30min for 64%(P<0.05), while the change rate of ADO level in control with moxibustion group increased 86%at 60min(P<0.05).There were no differences among other inter-groups at each time point (P> 0.05).2. The changes of local purines under pathological condition1) The change rate of ATP level in CFA with acupuncture-rotating group reached a peak at 30min with 76% and began to decrease from 60min when compared with CFA group(.P<0.05),whereas the ATP level was much higher. The change rate of ATP level in CFA with moxibustion rised at 30min with 28% and 16% at 60min(P<0.05).The change rate of ATP level in CFA with acupuncture-rotating group were significantly higher than that in CFA with moxibustion group at all the time points(P<0.05),as well as in acupuncture-without-rotating group at 30min(P<0.05). There were no differences among other inter-groups at each time point (P> 0.05).2) The change rate of ADO level in CFA with acupuncture-rotating group were significantly higher than that in model group at 30min(195%) and 60min(76%)(P<0.05),and CFA with acupuncture-without-rotating group were at 30min(142%) and 60min(110%), while the change rate of ATP level in CFA with moxibustion group increased 104% at 90min when compared with model group (P<0.05).The change rate of ADO level in CFA with acupuncture-rotating group were significantly higher than that in CFA with moxibustion group at 30min(P<0.05). There were no differences among other inter-groups at the remaining time points (P> 0.05)Conclusions:1.There is a difference of time response of local purines when stimulated with acupuncture and moxibustion under physiological condition.The ATP and ADO level were elevated at 30min and maintained with 30min by needle rotating.However, ATP and ADO level under moxibustion displayed a delayed elevation at 60min.2. There is a difference of amplitude response for local purines when stimulated with acupuncture and moxibustion under pathological condition.The ATP and ADO level were elevated at 30min both with needle rotating and moxibustion.However, acupuncture can last another 30min.Needle rotating raised more ATP and ADO than moxibustion and acupuncture without rotating.
