Object:This experiment through the observation with the activating blood components of ShenQi Compound Recipe,which has the effect of nourishing yin, tonifying Qi and activating blood, regulating endoplasmic reticulum stress of KKAy mice to keep the internal environment stability of organism which has diabetes vascular lesions, preliminary discussion the mechanism of the early application of Activating blood to prevent and treat diabetic vascular lesions,providing molecular theory basis for the importance and scientific of activating blood method in the prevention and treatment of diabetes vascular lesions of the of early intervention.Methods:Experiment with 50 7-8 weeks male KKAy mices,with random blood glucose greater tendency for 13.9mmol L/1 for 2 times and were randomly divided into 5 groups:blank group,model group,ShenQi Compound Recipe, activating blood grooup,atorvastat in group,each group of ten only.After adptive feed 1 week, blank group set to be sterilized water and normal feed,the rest of the group are joining nitric oxide synthase inhibitors (L-NAME) of 0.2 mg/ml/d in drinking water,at the same time giving high fat feed to make mode,building cycle is 35 days,diabetes atherosclerosis model was established.Building while giving drugs,the blank group,model group are calculated by filling 10ml/kg/d physiological saline;ginseng and astragalus compound group by taking 14.4g/kg/d ginseng and astragalus compound extract irrigateion,activating blood group according to take 2.7g/kg/d irrigation of ginseng and astragalus with promoting blood circulation to remove blood stasis efficacy components in compound extract,atorvastatin group according to take 3.2g/kg/d irrigation of atorvastat in suspension.During the daily observation we should observe the general situation of animals,involve drinking and food intake, and weigh the weight each week.After five weeks of the experiment, put to death in mice,drawing materials to test limosis blood glucose, blood lipid and serum hypersensitive creactive protein in mice,take abdomen aorta of small rice for pathological section,after HE staining,observing pathological morphology changes under light microscope,using gene chip detection technology for gene expression spectrum detection of thoracic aorta tissue,extracting he endoplasmic reticulumt stress related genes,Statistical analysising and calculating in whether there is a difference between groups,to determine the pathway is activated or suppressed.Results:ShenQi Compound Recipe,activating blood group of general status in mice were better than the model group,the blood lipid,hs-CRP were lower than the model group,the improvement of ShenQi compound Recipe is better than that of blood group.Compared with the blank group,the expression states of gene Hspa5, Casp12 in model group are increased,the expreesion of gene of Bcl2 and XIAP are down-regulated.After treatment with activating blood group,the expression states of gene Hspa5, Casp12 are downregulated,the expression of gene of Bcl2 and XIAP are increased.Conclusion:To improve the lipid metabolic disorder,reduce vascular endothelial injury,regulate the expression of endoplasmic reticulum stress related genes,to maintain or restore the endoplasmic reticulum steady state,may be ShenQi Compound recipe early application of blood components delay or prevent the progres of atherosclerosis,thus the mechanism of action of prevention and cure of diabetes vascular lesions.
