Torreya grandis ‘Merrillii' is a species of conifer in either the family Taxaceae,or the Torreya.Torreya grandis ‘Merrillii' is selected from the Torreya grandis Fort.ex Lindl due to its variance by artificial selection,and it is chosen as the material for cultivating and grafting.Generally,it is common that grafting is used during the plantation of Torreya grandis ‘Merrillii'.However,it takes a multitude of time about the period of growth and the healing for the cutting of grafting seedlings in the traditional way,which uses 2+2 seedlings as the materials.In this situation,micrografting can solve these problems and boost the reproduction coefficient.In the micrografting process,the scion and stock are chosen from materials which are non-lignified and semi-lignified.The time of healing is only 10 days,and it can supply the good seedlings of Torreya grandis ‘Merrillii' in short time.On the other hand,doing some research on grafting and the healing growth can make a foundation for developing the technique of micrografting and industrialization.In this paper,based on the technique of micrografting,researching on the anatomical observation and the measurement of physiological parameters during the healing process.The result shows that:(1)The average germination of seeding stand of Torreya grandis ‘Merrillii' is 64.0% after seed stratification in 3 months.Its rate of seed germination and mildew are 32.0% and 1.7% which broke the dormancy stored in 4 ? wet sand in fridge are less than 37.7% and 12.3% of the bag to hide.The germination rate of sowing is 95.0% in October after two months,and the period of seed stratification is about 15 days,and it is used as stock when grows in 15 days.(2)The 3 cm length of stock and scion,3mg/kg' IAA is the best for the survival by used cleft grafting which have the better grafting survival rate and seedling rate of Torreya grandis grafting.(3)Cleft grafting is better than drilling and side grafting in the healing and growth of Torreya grandis ‘Merrillii',which healing within 15 days.The survival rate of cleft grafting is much more than that of drilling grafting and side grafting,which is 66% and 68% respectively.Scion which do not have top bud grows better than that has top bud.The parameter among new stubble,diameter and seedling height have the significant correlation.It has the same situation in the germination and growth period.Micrografting can be done all the year,where the survival rate of the graft in June and July are 90.0%,and the rate of seedling is above 83.3 percent.In April,the graft was worse.(4)During the observation of healing process,it can be seen that the key is the callus tissue of scion and stock.It forms a isolation layer after grafting in 2 days and the callus tissue comes out in 4 days.In the next 4 days,the callus of scion and stock meets and then differentiation.When it is 10 days,it has been healed and has the ability to transport.(5)The parameter of POD,CAT and soluble protein of Torreya grandis ‘Merrillii' in breeding box is much better than in incubator.In breeding box,the POD reaches the lowest in 2 days and then gradually increase in the next 6 days;the CAT reaches the peak in 6 days;soluble protein has the same situation correlation to POD.correlation analysis revealed that soluble protein has a positive correlation with POD not only in scion but also in stock,while the soluble protein has a negative correlation with CAT.The activity of POD is made by the connection between scion and leaves.The amount of soluble protein has a positive correlation between the scion and graft union.
