Devepment Of Blocking ELISA Kit For Detection Of The Antibody Against Porcine Circovirus Type 2 And Comparison Of Its Effectiveness With Commercial ELISA Kits

Porcine circovirus(PCV)is the smalliest animal virus that has two serotypes,PCV1 and PCV2,but in which only PCV2 has pathogenicity.PCV2 can damage immune organ inducing immunosuppression which make secondary mixed infection and resulting in porcine circovirus associate diseases(PCVD),such as postweaning multisystemic wasting syndrome(PMWS),porcine dermatitis and nephropathy syndrome(PDNS).And PMWS has caused great economy loss in swine industry.The Capsid is the major structure protein of PCV2 and it had many B cells epitopes and neutralisation epitopes,which can induce immunoprotection in pigs.The diagnostic techniques of PCVD include clinical observation,histopathology observation,antigen detection and antibody detection.Among them,antibody detection method include ELISA,IPMA,IFA.They are used to detect mass clinical serum since result assessment can be standardization.Our laboratories had established blocking ELISA antibody detection method based on the HRP labeled 5H7 monoclonal antibody for its high specificity and had conservation epitopes to bind capsid protein.It also could be used to evaluate the efficiency of vaccine immunity.In this study,based on PCV2 Cap protein 5 baths of the PCV2 blocking ELISA antibody detection kits were manufactured in Nannong Hi-Tec Company GMP workshop by preparing PCV2 Cap recombinant proteins,HRP-McAb and the positive and negative serum antibodies to PCV2.And the trait test,sterling test,susceptibility and specificity were detected and evaluated,respectively.The results showed that quality of those kits e is consisting to the quality standard of the product.Meanwhile,Total 598 swine sera samples were collected from five pig farms in Jiangsu province,and our kits and two commercial ELISA kits were used to detect the antibodies to PCV2 made by Ruipu LTD in China and Jinnuo Company in South Korean.The results showed that the coincidence rates of our kits with kit made by Ruipu and Jinnuo Company were 81.8-84.5%and 93.6-94.5%,respectively.The results indicated that the PCV2 blocking ELISA antibody detection kits can be used for PCV2 antibody detection in swine.
