Rice(Oryza sativa L.)accounts for about 40%of China’s total grain,which is the most important food crops in the world.Because the rice genome sequencing has been completed,it has become a typical model of crop plant genomics studies.Transgenic rice is widely used in breeding or gene functional analysis by Agrobacterium mediated method,the advantages of this method is high efficiency,low cost,short cycle,copy number and low gene silencing phenomenon less etc.There are close relation between the rice plant regeneration and transformation rate of varieties and their genetic differences.Now,the transgenic rice research was gradually focus to how to establish a new type of callus regeneration system and Agrobacterium transformation system.In this study the mature emberyo and immature embryo of japonica rice varieties,Ningjing No.1,Nipponbare,Donjing was used to transform by Agrobacterium tumefaciens mediated method。The order of this study was build a set of efficient transformation system by Agrobacterium mediated in rice.The results show that,the embryogenic callus induction rate in immature embryos of different varieties exist differences.Compared with Ningjing No.1 and Nipponbare,the mature embryo and immature embryo callus induction rate in Donjing were highest,reach to 96.2%and 96%,respectively.The embryogenic rate also was high to 95%.While,the transformation rate of target gene was different among the three varieties.The resistant callus rate and resistant plant conversion rate was the highest in Nipponbare mature embryos callus and reach to 46.2%and 45.6%,respectively,and its possible causes are discussed.
