The heterosis utilization of soybean is an important field of current research,but also an effective way to improve the mining potential yield of soybean,soybean production.At present,China's soybean heterosis and its utilization in the international leading position,highly efficient hybrid soybean seed production technology,strong heterosis breeding,hybrid certification,have made considerable progress.In order to further improve the utilization of heterosis potential,still need to expand the sterile line and restore line breeding range and speed up the breeding,this will request in cytoplasmic male sterile line of mode of inheritance and molecular marker assisted selection in the fundamental areas of in-depth inquiry.In this study using soybean cytoplasmic male sterile lines SXCMS1A and restorer AHH-8 to build F1 hybrid populations,F2 segregating population fertility,to determine plant fertility by microscopy combined with mature plants pollen fertility performance,plants obtained fertility segregation.According to F1,F2 group of the separation of the male sterile gene with restorer gene number and recessive relationship discussion,clear the genetic pattern of soybean cytoplasmic male sterile line SXCMS1A;Using 445 SSR primers of soybean to fertile gene pool,sterile gene pool,half male sterility gene pool has been screened polymorphism,the polymorphic primers were used to verify the single plant in the F2 population,and the gene was preliminarily located.The following results were obtained:1.Soybean cytoplasmic male sterile lines SXCMS1A as female parent and to restore the AHH-8 as male parent of hybrid,the F1 hybrid populations,F2 population.Through pollen microscopic examination,and combined with the mature period of field Fertility Survey judging plant fertility.The F1 population of 306 plants all showed semi sterility,the F2 population of 293 plants can be breeding lines and semi sterility were 139 strains,154 strains,by chi square for test to detect segregation ratio fits 1:1.The soybean cytoplasmic male sterile line SXCMS1A is gametophyte sterility typical single gene control.2.Method to determine plant fertility experiment in the pollen fertility rate of microscopy and observed in mature period per plant type characteristics combined can effectively reduce due to insufficient number of buds of acquisition caused by microscopic examination of error,at the same time,it can avoid the insect pollination resulted in some sterile strains of a large number of pod,therefore,it is able to avoid the error of fertility judgment.And then affect the accuracy of the experiment.Analysis of various fertility plants in the study of genetic model,The study found that the expression of male sterility in soybean was regulated by environmental conditions.3.In the mode of genetic research based on selected 445 of soybean SSR primers of F2 segregation population for SSR marker analysis,is obtained by the D1A linkage groups 2 and restoring genes tightly linked markers satt184 according to and Satt267.The genetic distance were 17.3cM and 10.2cM,Genetic mapping was carried out using Map Join 4 software,and the gene was located on D1a linkage group.
