The rate of isolation of Umbilical Cord Mesenchymal Stem Cells(UC-MSCs)and quanity of UC-MSCs after isolation will decrease,even lack of stemness if umbilical cord kept in 4 degree celsius or minor 20 degree celsius for long time.This will cause more cost of acquisition,production and testing.The article will introduce the optimization of storage of UC-MSCs.In the frozen liquid,using three-steps cooling method to keep the umbilical cord tissue which simply processed from freshly collected umbilical cord into the liquid nitrogen(-196?).Tissue can be recovered in anytime for production.The tissue block adherent method of isolation of UC-MSCs can be 100% sucessful and all the test indicators meet the production requirement.The technician of keeping fresh umlibical cord in liquid nitrogen reduce the high umbilical scrap rate to zero and sovle the supply problem in inadequate acquisition period of umbilical cord.To select therapy of producton of the umbilical cord mesenchymal stem cells injection,lower the cost,using 0.9% Sodium Chloride Injection,5% Dextrose Injection and Compound Electrolytes Injection in different time and temperature to study the effection of survival rate,adherent ability and doubling time,finding that the Compound Electrolytes Injection is surpass the other two for keeping the biological activity of UC-MSCs.Meanwhile,in order to reduce production costs and allergic reactions during infusion incidence of compound electrolytes containing different concentrations of human serum albumin injection to save the biological activity of UC-MSCs were compared and found that 1% of the human blood albumin that can significantly improve the survival rate of cells.At 4? condition storage for 24 hours,cell viability,surface-specific antibodies,multi-directional differentiation,immune regulation,growth and proliferation,such as testing indicators to fully meet FDA requirements for active cell transplantation,and in line with the company's quality standards.Then saving about 50% of the amount of human serum albumin and reducing the indicence of human serum albumin allegic reactions.
