An Experimental Study Of The Related Mechanism Of Jiuxiang Zhixie Tablets In The Prevention And Treatment Of Damp-heat Type Acute Infectious Diarrhea

[Objective] The purpose of the present study was to investigate the mechanism of Jiuxiang Zhixie Tablets (also known as Chin Xiang Zhixie Tablets) on common intestinal bacteria by observation the effects of Jiuxiang Zhixie Tablets (also known as Chin Xiang Zhixie Tablets) on mouse serum IL-8 level, hematological index leucocyte and lymphocyte count, and in vitro and in vivo bacteriostasis experiments. We further explored its role in the treatment of damp-heat diarrhea, in order to provide more scientifically theoretical and experimental evidence for its clinical application.[Methods] Folium sennae was used to set up the animal model and the effects of high, medium and low dosage Jiuxiang Zhixie Tablets (Jiuxiang high, medium and low dosage) on diarrhea index were compared with Changkang Tablets, Belladonna Sulfamethoxazole and Trimerhoprim Tablets.Two-fold dilution test tube method was used for determination of minimum inhibitory concentration (MIC), and cup-plate method was used for the determination of the minimum bactericidal concentration (MBC). The bacteriostasis of Jiuxiang Zhixie Tablets was compared with related drugs. Diarrhea infection animal models were established by intraperitoneal injection of three different bacteria suspensions containing lethal dose of E. coli, bacterium paratyphosum B and Shigella bacteria in mice, respectively. The protective effect and effects on survival time of high, medium and low doses of Jiuxiang Zhixie tablets (hereinafter referred to Jiuxiang high, medium and low dose group), Changkang Tablets and Norfloxacin in mice infection model were observed. Blood was collected through the heart before the animals were killed by aseptic processing, blood IL-8 contents was measured by double antibody sandwich enzyme linked immunosorbent assay (ELISA), and mouse leukocyte and lymphocyte count were performed through Wright stain.[Result] Diarrhea index was reduced in Jiuxiangzhixie Tablet groups and the effects was superior to Changkang Tablets, Belladonna Sulfamethoxazole and Trimerhoprim Tablets (p<0.05). The MIC of Jiuxiangzhixie Tablet against Escherichia coli, Staphylococcus aureus, Bacterium paratyphosum B, Shigella dysenteriae and bacillus proteus were 0.25g/mL、0.125g/mL、0.25g/mL、0.25g/mL、0.25g/mL respectively; The MBC of Jiuxiangzhixie Tablet against Escherichia coli, Staphylococcus aureus, Bacterium paratyphosum B, Shigella dysenteriae and bacillus proteus were 0.25g/mL 0.125g/mL、0.25g/mL、0.5g/mL、0.5g/mL respectively. Jiuxiangzhixie Tablet had a significantly better bacteriostatic action against Escherichia coli and Shigella dysenteriae compared with Changkang Tablet (p<0.05). All drug treatments significantly increased the survival rate of infected mice injected with Escherichia coli, Bacterium paratyphosum B, and Shigella dysenteriae, and prolonged the survival time (p all< 0.05) Compared with Changkang Tablets group, norfloxacin group and Jiuxiang high dose group had better effects (p all<0.05). Jiuxiang medium dose group was superior to Changkang Tablets group; the extension of survival time in Jiuxiang high dose group was comparable with norfloxacin group; In Jiuxiang Zhixie Tablets groups, the protective effect of Jiuxiang Zhixie Tablets on infected mice and survival time had a certain dose-effect relationship. Compared with infection model, Jiuxiang groups significantly reduced inflammatory factors IL-8 contents, leucocyte count and increased lymphocyte count (p all<0.05). The difference between Jiuxiang high dose group and Changkang Tablets was significant (p<0.05).[Conclusion] The results suggest that Jiuxiangzhixie Tablets had an ideal effects to stop diarrhea and inhibitory action on bacteria in vitro and had a good protective effect on mouse models that received intraperitoneal injection of lethal dose of Escherichia coli, Shigella dysenteriae and Bacterium paratyphosum B. Jiuxiangzhixie Tablets could significantly improve protection rate, reduce inflammatory factors IL-8 contents and leucocyte count, increase lymphocyte count, and significantly prolong the survival time of infected mice, showing a dose-effect relationship.
