ObjectiveShuanghua granules is a herbal products developed by use the guidance of the TCM theory by our team which made by L.macranthoides hand and Dendranthema morifolium (Ramat.) Tzvel. It can clearing away the heat-evil and expelling superficial evils and good for nourish body in hot summer. L.macranthoides hand named da-ye honeysuckle also, is woody vines plants of caprifoliaceae endure genera whose flower bud and new flowers in Chongqing, Sichuan, Guizhou and Yunnan provinces used as honeysuckle.The objective of this essay is research on the pharmacological activity of Shuanghua granules in vitro bacteriostasis, anti-inflammatory and immunoregulation.MethodsIn vitro bacteriostasis researchDisk diffusion method and 96-well plate microporous dilution method were applied to assay the MIC of the following bacteria:Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Bacillus aeruginosus, Arthrobacter luteus Kaneko and Pseudomonad.Anti-inflammatory action research An acute inflammation reaction model employing xylene as inflammagen to induce mouse ear edema were set up,50 KM mice divided into 5 groups randomly,to test anti-inflammatory activity of each group. They are model control group, positive control group (aspirin 0.2 g/kg), Shuanghua granules low doses middle dose and high group (2.25 g/kg,4.5 g/kg,9.0 g/kg). Calculate the inhibition rate.An acute inflammation reaction model employing carrageenan as inflammagen to induce rat paw swelling were set up,50 rats divided into 5 groups randomly,to test anti-inflammatory activity of each group. They are model control group, positive control group (aspirin 0.2 g/kg), Shuanghua granules low doses middle dose and high group (1.125 g/kg,2.25 g/kg,4.5 g/kg). Calculate the inhibition rate.A chronic inflammation model placeing cotton into abdominal cavity of rats to induced granuloma were set up,50 rats divided into 5 groups randomly,to test anti-inflammatory activity of each group. They are model control group, positive control group (aspirin 0.2 g/kg), Shuanghua granules low doses middle dose and high group (1.125 g/kg,2.25 g/kg,4.5 g/kg). Calculate the inhibition rate.Immunoregulatory actionThe immunosuppressed mice were induced by cyclophosphamide (Cy, ip).Index of immune organs was weighed and calculated. Phagocytosis of mononuclear macrophage was determined by cleaning carbon particle method. Index of K and a were calculated. Peritoneal macrophage proliferation and splenocyte lymphocyte transformation test of mouse was measured by MTT assay.ResultsIn vitro antibacterial experimentThe MIC and MBC of Staphylococcus aureus are 40 mg/mL and 1280 mg/mL, Escherichia coli 80 g/mL and 1280 g/mL, Micrococcus luteus 80 mg/mL and 1280 mg/mL, Bacillus aeruginosus 320 mg/mL and 1280 mg/mL, Bacillus subtilis 160 mg/mL and 640 mg/mL, Pseudomonad 320 mg/mL and 1280 mg/mL.Anti-inflammatory activitiesDifferent groups of Shuanghua granules have anti-inflammation effects obviously. Inhibition rate of mouse ear edema, low dose group 36.73%, middle dose group 66.05%,high dose group 68.52%. Inhibition rate of rat paw swelling are 18.97% to 68.99%.Inhibition rate of granuloma are 10.36% 17.67% 22.88% respectively for different dose groups.The influence of immunoregulatory miceCompare with model group, different dose groups all can increase the index of thymus and spleen (P<0.05). Thymus index in the mid-dosage group and high-dosage groups and increased significantly (9.60±0.59 mg/10g and 10.47±0.57 mg/10g). Spleen index in low middle and high doses groups were higher than model group significantly (14.51±0.89 mg/10g,15.69±0.36 mg/10g and 14.46±0.96 mg/10g).Compare with model group, different dose groups all can increase the index of K and a number (P<0.05). Index of K and a number in high dose group were higher than the model group significantly ((0.56±0.07)×10-2 and 3.86±0.79). Index of a number in middle dose group was higher than the model group significantly (3.66±0.31).The influence of mice cellular immuneCompare with blank group(0.024±0.005), low doses group (0.015±0.003) was no significant difference,middle dose group (0.042±0.006) and high dose group (0.043±0.005) were significant difference (P<0.05).The influence of mice abdominal macrophage proliferationCompare with blank group(0.087±0.003), low doses group (0.077±0.004) and middle dose group (0.101±0.002) were no significant difference, high dose group (0.173±0.005) and positive group (0.183±0.014) were significant difference (P<0.05).ConclusionShuanghua granules is antimicrobial significant, useful in inhibition chronic and acute inflammation,and has effects of cellular immune, monocyte phagocytosis and macrophage phagocytosis, which can enhance the immune function. All of these suggest that Shuanghua granules has a good development prospect.
