In order to isolate the active constituents in Chuanlongshuyu,various chromatographic techniques were performed.Five compounds were isolated and identified with IR,NMR and MS data and physical-chemical properties.They were dioscin,protodioscin,trillin methylprotodioscin and gracillin.A RP-HPLC method had been established for the fingerprint analysis of Dioscorea.The separation was achieved on a Shim-pack VP-ODS column(250 mm×4.6 mm,5μm)using acetonitrile-water as the mobile phase,208 mn as the detection wave-length.The 4 peaks were identified.Protodioscin was used as internal standard.The results of cluster analysis and similarity calculations of the raw materials showed the difference of collected samples.This method is simple,accuracy and stable for the establishment of quality control of Dioscorea.A simple,sensitive and specific HPLC method had been developed and validated for the determination of dioscin,protodioscin and trillin in Dioscorea.The method was validated over a linear range of 20.16~504.0μg·mL-1(r = 0.9989)of dioscin,10.28~514.0μg·mL-1(r = 0.9999)ofprotodioscin and 10.16~508.0μg·mL-1(r = 0.9998)of methylprotodioscin.The average recovery of dioscin was 96.2%with RSD of 1.0%(n = 9).The average recovery was of protodioscin 96.3%with RSD of 0.8%(n = 9).The average recovery of methylprotodioscin was 99.4%with RSD of 1.2%(n = 9).The developed method was reasonable and feasible to describe and evaluate the quality of Dioscorea based on the chemical data.We first established a method for the fingerprint analysis of Dioscorea by MEKC.The separation was achieved on a uncovered fused-silica capillary column(250 mm×4.6 mm,5μm) using sodinm biborate-SDS(25 mmol·L-1-10 mmol·L-1)as the buffer,208 nm as the detection wave-length.Protodioscin was used as internal standard,17 peaks were considered as common peaks in collected samples.Similarity calculations were studied by comparing the fingerprint chromatograrns of the Dioscorea and mutual pattern.This method is simple,accuracy and stable for the establishment of quality control of Dioscorea.Conclusively,the quality assessment methods of Dioscorea was developed,which provided a basis for the establishment of quality control.
