Pass One Comparison of cell suspension method and tumor tissue embe dding method in establishment of VX2 muscle tumor model in rabbitObjective:To compare the cell suspension method and tumor tissue embedding method in establishment of VX2 muscle tumor model in rabbit.Methods:After separating the VX2 tumor from cancer bearing VX2 rabbit, the tumor tissue blocks and cell suspension were prepared; the tissue blocks was embedded into the left vastus lateralis(tissue block group) and the cell suspension was injected into the right vastus lateralis(cell suspension group) of 10 rabbits. Color Doppler ultrasound was used to observe the tumor and measure the tumor volume 7, 14 and 21 d after model establishment. The rabbits were sacrificed and the tumors were taken out for volume measurement 21 d after model establishment.Result:The success rates were both 100% for two methods. The tumor volume in tissue block group was significantly greater than that in cell suspension group 7, 14 and 21 d after model establishment [(0.13±0.07) cm3 vs(0.05±0.01) cm3,(1.66±0.59) cm3 vs(0.94±0.30) cm3,(13.30±4.68) cm3 vs(7.15±2.65) cm3](P<0.05). Twenty one days after model establishment, the tumor sharp was regular and no ectopic implantation was observed in tissue block group, while the tumor sharp was irregular and ectopic implantation was observed in cell suspension group; compared with actual tumor volume, the volume measured by ultrasound show no significant difference [tissue block group:(13.30±4.68) cm3 vs(12.2±4.86) cm3, cell suspension group:(7.15±2.65) cm3 vs(6.42±2.33) cm3](P>0.05).Conclusions:The tissue embedding method was more convenient with less ectopic implantation in establishment of muscle tumor model compared with cell suspension method. Pass Two Application of Improved Biopsy Technique in Reducing Complications Induced by VX2 Tumor PunctureObjective:1 To establish the VX2 muscle tumor model of New Zealand rabbit and investigate the efficacy between the traditional biopsy techniques and the improved biopsy method in order to prove the improved biopsy method can improve local bleeding and tumor spreading.2 To explore the application value of the improved biopsy method in reducing complications in the limp tumors.Methods:The VX2 tumor was transplanted in thigh vastus lateralis of 22 rabbits using tissue mass embedding method to establish model of muscle.20 cases of adult rabbits were punctured. All the left muscle tumors were punctured with the improved biopsy method as the left group,which the puncture holes were implanted the bolt closure through the casing after the biopsy. And all the right tumors were punctured as the right group,which used traditional biopsy technique.The other two rabbits were the control group. Color Doppler Ultrasound was used to assess the bleeding of the puncture hole after 1 hour. The pathological study was carried out for the left and right group after 7 days,and the other two rabbits was carried out the pathological study after the VX2 tumor was transplanted for 3 and 4 weeks.Result:1 1 hour later,only 8 cases from the left group showed some degrees of exudation around the puncture hole, but there were 17 cases that showed some degrees of exudation around the puncture holes of all the 20 cases of the right group. The left group was lower than that of right group [(40%,8/20) vs(85%,17/20)], the difference was statistically significant(P<0.05). The hematoma volume of the left group was lower than that of right group [(032)mm3vs300(162) mm3], the difference was statistically significant(T=161, P<0.05).2 The pathological study was carried out after 7 days,there were 4 cases that we can find the tumor cells in the soft tissue near around the puncture hole in the left group, but 14 cases in the right group, but none in the control group. The left group was lower than that of right group [(20%,4/20)vs(70%,14/20)], the difference was statistically significant(χ2=10.10,P<0.05).Conclusions:1 Compared with the traditional biopsy technique, the improved biopsy method could improve the bleeding in the puncture hole.2 Compared with the traditional biopsy technique, the improved biopsy method could reduce the risk for tumor cell metastasis in the adjacent soft tissues.3 The improved biopsy method could reduce the complications Induced by VX2 Tumor Puncture, which is a check method and train of thought for the display and diagnosis of the tumor.
