| Objective:To observe primary cilia of epithelial cells at mouse embryonic palate by immunofluorescence and electron microscope.To lay foundation for exploring the relation between primary cilia and the transduction of shh signaling during the process of the development of mouse embryonic palate.Methods: 1.8 pregnant mice were collected and killed,and the mouse embryos were collect.Scanning electron microscope specimens with the mouse embryos were made and the structure of cilia of epithelial cells at mouse embryonic palate by SEM were observed. 2.8 pregnant C57BL/6J mice were collected and killed,and the mouse embryos were collected.Transmission electron microscope specimens with the mouse embryos were made and the structure of primary cilia of epithelial cells at mouse embryonic palate by TEM were observed.3.8 pregnant C57BL/6J mice were collected and killed,and the mouse embryos were collected.Paraffin sections were made with the mouse embryos.The primary cilia of epithelial cells at mouse embryonic palate by LSCM were observed and identified.Results: 1.At ED12.5/13.5/ED14.5,the results photographed by SEM showed that most of the epithelial cells at mouse embryonic palate were bulging and had a clear edge among adjacent cells,many epithelial cells extended cilia from the surface and the ciliary membrane was intact and smooth.Besides these,there was a large number of filopodia on the epithelial cells surface.At ED15.5,the mouse embryonic palates of mice had fused mainly.There was no cilia projecting from the cell surface and less filopodia projecting from the cell surface than ED14.5.2.The results photographed by TEM showed that primary cilia of various lengths projected from the cell surface of the epithelial cells at mouse embryonic palate at ED12.5/13.5/14.5,the basal body,the ciliary membrane and the cilia substrate were clear.Other cellular organelles were clear.Among these,theninefold symmetry axonemal structure of primary cilia were clear at ED12.5.Besides primary cilia,there was a large number of filopodia on the epithelial cells surface.But at ED15.5,the mouse embryonic palates had fused mainly.There was no cilia projecting from the cell surface of the epithelial cells of the oral side of mouse embryonic palate, filopodia projecting from the cell surface of part of the epithelial cells and other cellular organelles were clear.3.The results photographed by LSCM showed that most primary cilia located in the medial edge epithelial cells at ED12.5/13.5/ED14.5,and there’s no primary cilia of the epithelial cells at ED15.5.Conclusion: The experiment confirmed that primary cilia profect from epithelial cells at mouse embryonic palate at certain gestation and the morphology and structure of primary cilia is similar to those of other cells.With mouse embryonic palates fusing,the primary cilia of epithelial cells at mouse embryonic palate disappeared.It demonstrated that there is a certain relation between primary cilia and the fusion of mouse embryonic palates and laid foundation for exploring the relation between primary cilia and the key signaling transduction during the process of the development of mouse embryonic palate further. |
