| Objective:To investigate the effect of Tripterygium Wilfordii Polyglycosidium(TWP) on expresssion of in atopic dermatitis(AD), understand molecular immunology machanism of AD by TWP, and elucidate pathogenesy of molecular machanism in AD, and to provid some foundation about more rational use of the to treat AD.Methods:Separate peripheral blood monouclear cells(PBMC)from AD, and cultured in vitro. Determine the expression of IL-2ã€IL-4ã€IFN-γ and TNF-α by RT-PCR.1 By using the Ficoll-Hypaque density centrifugation to separate PBMC from AD, and cultured in vitro in different concentration of TWP for 48 hours. The drug level in the control groups, TWP groups were 0, 0.5, 1and 5mg/L.2 The expression of IL-2ã€IL-4ã€IFN-γ and TNF-α in PBMC in vitro treated by TWP were detected by RT-PCR.Results:1 The TWP on expression of IL-2 m RNA in in AD, except the treatment group of 5.0mg/L had significante statistics compared with that in the 0.5mg/L group, and that in the control group(P<0.01 or P<0.05);expression shown statistical significance between the 1.0mg/L group and control group the group of 0.5mg/L(P<0.01); however the rest group comparison had no statistical significance(P>0.05).2 The effect of TWP on expression of IL-4 mRNA in AD, except 5.0mg/L group and the 1.0mg/L group had no statistical significance( P > 0.05), the rest group comparison had statistical significance(P<0.01 or <0.05).The expression level of IL-4 of PBMC of patients with AD was more higher than that of normal control(P<0.01).3 The effect of TWP on expression of IFN-γ m RNA of patients with AD, except 5.0mg/L group and 1.0mg/L group, 1.0mg/L group and 0.5mg/L group,0.5mg/L group and control group had no statistical significance(P>0.05), the rest group comparison had statistical significance(P<0.01 or <0.05).The expression level of IFN-γ of PBMC of patients with AD was lower than that of normal control/L.4 The effect of TWP on expression of TNF-α m RNA in AD, except 5.0mg/L group and 1.0mg/L group, 1.0mg/L group and 0.5mg/L group had no statistical significance(P>0.05), the rest group comparison had statistical significance(P<0.01 or <0.05).The expression level of TNF-α of PBMC of patients with AD was lower than that of normal control.Conclusion:1 The expression of level of IL-4 mRNA of patients with AD was more higher than that of normal control, however that of IFN-γ m RNA of patients with AD was lower than of normal control, so there was immune unbalance of Th1/Th2 in AD.TWP could inhibit the expression of IL-4 and IFN-γ at different extent,so reduced the expression level of two, ameliorating condition of unbalance of Th1/Th2.2 TWP could inhibit the expression of IL-2 of PBMC cultured in vitro of patients of AD. And at 1.0mg/L and 5.0mg/L, the effcet was significant. TWP could inhibit the proliferation of T, B cells by inhibiting expression of IL-2, thereby it would affect expression of lots of cytokines to educe immune accommodation.3 The expression level of TNF-α of PBMC of patients with AD was lower than of normal control.4 TWP at 1.0mg/L and 0.5mg/L, could educe immunosuppression well. |
