| PART 1 The expression of mediators of inflammation in the rat model for nonbacterial chronic prostatitisObjection: In our present research we adapted nonbacterial chronic prostatitis model to research the expression of TNF-α,ICAM-1,NF-κB,iNOS in the rat model of nonbacterial chronic prostatitis, and study the role of TNF-α,ICAM-1,NF-κB,iNOS in the rat model of nonbacterial chronic prostatitis.Method: 30 1-year-old male Sprague-Dawley rats were divided into two groups randomly, the normal group and the model group. The normal group received no treatment. The model group was castrated and injected estradiol benzoate (0.25 mg/kg/day, s.c.) for 30 days. After treatment for 30d, The number of WBC and lecithin body in prostate tissue were measured,the pathologic appearance of the prostate tissue of the two groups were observed by HE staining and light microscope, and TNF-α,ICAM-1, NF-κB,iNOS in prostate were detected by means of immuneohistochemictry (sp).Results: The control group did not have inflammatory reaction in their prostates,while the model group displayed visible inflammatory reaction. There is Negative expression of TNF-α,ICAM-1,NF-κB,iNOS in normal group and Positive expression of TNF-α,ICAM-1,NF-κB,iNOS in the model group. The expression in normal group were lower than that of in model group(P<0.01).Conclusion: TNF-α,ICAM-1,NF-κB,iNOS play an emportent role in the development of the nonbacterial chronic prostatitis and maybe the new measurement for nonbacterial chronic prostatitis.PART 2 The effects of tripterygium wilfordii polyglycosidium in the rat model for nonbacterial chronic prostatitisObjection: In our present research we adapted nonbacterial chronic prostatitis model to study the effection of tripterygium wilfordii polyglycosidium in nonbacterial chronic prostatitis,and to investigate the possible mechanism,provide a new method in clinical treatment.Method: 30 1-year-old male Sprague-Dawley rats were divided into three groups randomly, the normal group, the model group, the TWP group. After the model finished, the TWP group was given tripterygium wilfordii polyglycosidium 10mg/kg, three times a day ,po ,and others were given the same normal sodium,po. After treatment for 7day, The number of WBC and lecithin body in prostate tissue were measured, the pathologic appearance of the prostate tissue of the three groups were observed by HE staining and light microscope and TNF-α,ICAM-1,NF-κB,iNOS in prostate were detected by means of immunohistochemictry(SP) .Results: The control group did not have inflammatory reaction in their prostates.the model group displayed visible inflammatory reaction. Inflammatory reaction remitted obviously in the TWP group. There is Negative expression of TNF-α, ICAM-1 , NF-κB , iNOS in normal group .The expression of TNF-α,ICAM-1,NF-κB,iNOS in normal group became significantly lower than the others group(P<0.01). The expression of TNF-α,ICAM-1,NF-κB,iNOS in the TWP group were lower than that of in the model group(P<0.01).Conclusion: Tripterygium wilfordii polyglycosidium can low down the expression of TNF-α,ICAM-1,NF-κB,iNOS in the rat model of nonbacterial chronic prostatitis and maybe a new treatment for the nonbacterial chronic prostatitis. |
