| Objective: To study of the role human epidermal growth factor receptor 2 and tetraspanin 1 protein expression in the occurrence and progression of gastric carcinoma.To discuss the relationship between HER2 and TSPAN1 protein the expressions with patients clinical and pathological characteristic in gastric carcinoma.Methods:The expression of HER2 and TSPAN1 protein was detected by two-step Envision immunohistochemical,in 64 cases of gastric carcinoma tissue and para-cancerous mucosa,20 cases of normal gastric mucosa specimens were used as the control.Results:The positive rate of the HER2 protein expression in gastric carcinoma tissue 23.4%(15/64) was significantly higher than that in normal gastric mucosa 0%(0/20) with statistical significance(P<0.05).The positive rate of the HER2 protein expression in para-carcinoma mucosa 10.9%(7/64) was higher than that in normal gastric mucosa 0%(0/20) without statistical significance(P>0.05).Meanwhile,the positive rate of the HER2 protein expression in gastric carcinoma tissue 23.4%(15/64) was higher than in para-carcinoma 10.9%(7/64) without statistical significance(P>0.05).The positive rate of the HER2 protein expression in intestinal-type gastric carcinoma with Lauren type 33.3%(14/42) was significantly higher than in diffuse-type or mixed-type gastric carcinoma 4.5%(1/22),(P<0.05).The expression of HER2 protein in the high differentiated gastric carcinoma 38.5%(5/13) and the middle differentiated gastric carcinoma 31.0%(9/29) was significantly higher than that in the poorly differentiated gastric carcinoma 4.5%(1/22),(P < 0.05). The expression of HER2 protein in the lymphatic metastasis gastric carcinoma 36.8%(14/38) was also significantly higher than that in the non-lymphatic metastasis gastric carcinoma 3.8%(1/26),(P<0.05).There was no significant difference among the expression of HER2 and the patient’s age,gender,tumor size,location,and TNM stage(P>0.05).The expression of TSPAN1 protein in gastric carcinoma tissue 68.8%(44/64) and para-carcinoma mucosa 23.4%(15/64) was significantly higher than that in normal gastric mucosa 0%(0/20),(P<0.05).The expression of TSPAN1 protein in gastric carcinoma tissue was significantly higher than in para-carcinoma(P<0.05).The expression of TSPAN1 protein in intestinal-type gastric carcinoma with Lauren type 52.4%(22/42) was significantly lower than in diffuse-type or mixed-type gastric carcinoma 100%(22/22),(P<0.05).The expression of TSPAN1 protein in the poorly differentiated gastric carcinoma 100%(22/22) and the middle differentiated gastric carcinoma 62.1%(18/29) was significantly higher than that in the high differentiated gastric carcinoma 30.8%(4/13),(P<0.05).There was no significant difference among the expression of HER2 and the patient’s age,gender,tumor size,location,metastasis and TNM stage(P>0.05).The expression of HER2 protein showed certain consistence with TSPAN1 protein(X2=7.107,r=0.349,P=0.003).Conclusion : The expression of HER2 and TSPAN1 protein is related with the occurrence and progression of gastric carcinoma,such as,differentiation degree,Lauren type,and the expression of HER2 is related with lymph node metastasis..The expression of HER2 and TSPAN1 protein isn’t related with age,gender,tumor size,location,and TNM stage,but,the expression of TSPAN1 isn’t related with lymph node metastasis. |
