| Objective: Vascular disease of low extremities(VDLE) is important causes of amputation and disability of T2 DM, its pathological basis is atherosclerosis(AS). Previous studies found that the AS closely related to lipid metabolism disordersã€oxidative stress, etc. LOX-1 is one of concerned AS therapeutic targets in recent years, it could through a variety of ways to promote the develop of AS. Studies have shown that acupuncture and moxibustion have effect on preventing and curing of VDLE, but the specific mechanism is unclear. After fed with high-fat diet and rhubarb, and injected streptozotocin(STZ) into the caudal vein, Qi deficiency and blood stasis T2 DM rat model is established. Acupuncture and moxibustion at pancreas and Zusanli acupoints, to observe the effect on the expression of blood lipid metabolism〠serum SODã€MDAã€Ox-LDL and LOX-1 of femoral artery.Methods: 38 SD male rats of 8 weeks feed for 1 week, were randomly divided into two groups: 8 control rats and 30 experimental rats. Control group were fed with common diet,while experimental group were high-fat diets for six weeks.Weight all of the rats on the next morning, experimental group were injected with STZ into the caudal vein as 25mg/kg,while control group were saline for comparison. After one week, take blood to measure blood glucose, experimental group that random blood glucose were higher than 16.7 mmol/L were successively T2 DM, lower than 16.7 mmol/L were injected with STZ again. Experimental group were divided to model group and acupuncture and moxibustion group randomly. Acupunture and moxibustion group take the acuponits of pancreas and Zusanli, electroacupuncture treatment was given by Yingdi KWD 808 I type pulse acupuncture therapy, 1 m A, 2 HZ, moxibustion at Zusanli,each 10 minutes, once a day, total 4 weeks. All of the rats were fasted the day before killed, under anesthesia collected blood sample to measure the expression of lipid fourã€serum SODã€MDA and Ox-LDL,taking the femoral artery HE staining to observe the pathological changes, examing the expression of LOX-1 by immunohistochemical staining, Western Blot to measure the protein expression of LOX-1.The data obtained was analysed by SPSS 16.0Results:1〠After fed with high-fat diet and injected STZ into the caudal vein, the achievement ratio of T2 DM model were 96.67%. 2〠the serum TCã€TGã€LDL-C of model group and acupuncture and moxibustion group were higher than control group(P<0.05), the serum TCã€TGã€LDL-C of acupuncture and moxibustion group were significant lower than control group(P<0.05),but the serum HDL-C was no significant difference(P>0.05). 3〠Femoral artery of model group occur early vascular disorders,while normal group and acupuncture group almost no pathological change. 4〠the serum Ox-LDL and MDA of model group were significant higher than control group(P<0.05), the serum Ox-LDL and MDA of acupuncture and moxibustion group were significant lower than control group(P<0.05). the serum SOD of model group and acupuncture and moxibustion group were significant lower than control group(P<0.05), the serum SOD of acupuncture group were significant higher than control group(P<0.05). 5〠The protein expression level of LOX-1 of model group was significant higher than control group, and the expression level of LOX-1 protein of acupuncture and moxibustion group is lower than model group(P<0.05).Conclusions: 1〠High-fat diet plus many low doses of tail-vein injection of STZ could successfully induced T2 DM rats model, the method is simple, the ratio is high. 2〠The expression of LOX-1 is higher in early VDLE rats, maybe related to the disorder of lipid metabolism, the higher expression of MDA and Ox-LDL and the lowerer expression of SOD. 3〠Acupuncture and moxibustion could reduce the expression of LOX-1,the mechanism may be related to regulate lipid mrtabolism,reduce the expression of MDA and Ox-LDL and increase the expression of SOD. 4〠Acupuncture and moxibustion could improve VDLE by reducing the expression of LOX-1ã€MDAã€Ox-LDL,and increasing the expression of SOD. |
