Background: The prescription Gubenfangxiao decoction (GBFXD) is an experiential effective recipe that used in clinical for many years.It can improve body congestion of fluid-retention by regulating lung, spleen. Preliminary pharmacological study shows it can alleviate the airway inflammation.This study based on congestion of fluid-retention equivalent to chronic airway inflammation in asthma.Chemokines are chemical attractant of the inflammatory cell.Whether GBFXD against asthma is involved in regulation chemokines.thereby reducing the inflammatory cells infiltration, play the role of airway inflammation in mitigation.Objective:To investigate the effect of GubenFangxiao Decoction (GBFXD) on the CCL11、 CCL20 in a murine modelMethods:42 Balb/C female mice were randomly divided into 7 groups with 6 rats in each group.The mice challenged by intraperitoneal injection of ovalbumin (OVA) Except the normal group, then changed by repeated inhalation of OVA and nasal inhalation of respiratory syncytial virus (RSV) to induced exacerbation of asthma remission in mice model.gbfxd daily gavage once for the first time of the prevention group sensitized, a total of 86 days. After the model was finished, the mice were divided into five groups in addition to the prevention group, the rats were gavage30 days except the model group, after the last administration beyond 24h, The effect of the drug on the expression of CCL 11 and CCL20 in the serum of mice was observed.Results:(1) Compared with the normal group, the average concentration of CCL 11 in serum of mice in model group is higher than that of the normal group, but no statistical significance (P> 0.05); compared with the model group, the average concentration of CCL 11 in serum of mice of prevention group and drug therapy group is lower than that of the model group (P <0.05). (2) Compared with the normal group, the average concentration of CCL20 in serum of mice in model group is higher than that of the normal group(P<0.05); compared with the model group, the average concentration of CCL20 in serum of mice of prevention group and drug therapy group is lower than that of the model group (P<0.05). (3) Compared with montelukast group, GBFXD low, medium and high dose group had no significant difference in the average of serum CCL11 (P>0.05); Montelukast sodium with Guben Fangxiao Decoction low dose group between CCL20 levels were significantly different (P<0.05), with the middle and high dose groups between CCL20 levels were not significantly different(P>0.05).Conclusion:(1) The mechanism of GBFXD treatment of asthma may be associated with CCL11,CCL20;and the relationship with CCL20 more closely. (2) Repeated attacks of asthma,early intervention and medication in clinical treatment is necessary. (3) The GBFXD alleviate the disease needs proper clinical dose to achieve better control.
