| OBJECTIVE:To investigate the effect of GubenFangxiao Decoction (GBFXD)on the airway inflammation and expression of asthma susceptibility gene ORMDL3in a murine model of Respiratory syncytial virus (RSV) induced asthma.METHODS:The asthma model was reproduced in the Balb/C mice challenged by repeated inhalation of ovalbumin(OVA) and repeated intranasal instillation of RSV (1.0×106.5TCID50/mL in50μL). After Balb/C mice’s administration of a dose (2.6mg/kg) of montelukast acetate, and three doses(12g/kg,24g/kg and36g/kg) of GBFXD, airway inflammation and expression of asthma susceptibility gene ORMDL3were observed. The airway inflammation was detected by lung histopathology and morphometry. Real-time quantitative polymerase chain reaction(RT-PCR) and Western blot assay were employed to confirm the expression of ORMDL3in the lungs of murine model at mRNA and protein levels, respectively.RESULTS:Compared with the control group, the expression of ORMDL3, TGF-p and IL-6were significantly higher, in the model group (P<0.05, P<0.01). Compared with the model group, GBFXD treatment significantly reduced airway jnflammation, and to some extent, inhibited collagen deposition and mucus production(P<0.05). RT-PCR and western blot assay indicated that GBFXD treatment downregulated the mRNA and protein expressions of ORMDL3respectively (P<0.05, P<0.01).However, the levels did not differ significantly between the GBFXD group and the montelukast group.CONCLUSION:The administration of GBFXD could inhibit the established airway inflammation and downregulated the expression of asthma susceptibility gene ORMDL3in allergic asthmatic mice,which may be related to multiple factors, including alleviation of airway inflammation,and inhibition of airway remodeling. |
