Using in vitro methods, for the purpose of being able to reasonably use Pinellia during pregnancy, prenatal and postnatal care and provide scientific, reliable experimental basis of pregnancy contraindication medicine research, the trial on the effects of Pinellia containing serum on mouse embryonic stem cell proliferation and differentiation was conducted by the Institute of Pediatrics of Nanjing University of Chinese Medicine.Objective1.To summary pregnancy drug contraindications significance in the history of development of Chinese medicine health science in children.2.To compare the impact on the proliferation and differentiation of mouse embryonic stem cells with different Raw Banxia (pinellia ternata)-containing serum compatibility.3.Explore the rationality of Ginger ginseng pinellia decoction (powder) was applied as pregnancy contraindication medicine.Methods1.Through the preparation of medicated serum in rats in mouse ESC, on first, third and fifth day research by applying the method of thiazole blue (determined by MTT) medicated serum influence on mouse embryos ESC proliferation, the statistical soft wares of SPSS 19.0 were used.2.Groups in cell apoptosis experiment was used to test the drug-containing serum on first, third and fifth day influence on mice ESC apoptosis, the statistical soft wares of SPSS 19.0 were used.3.Through cultivation mice ESC differentiation, using real-time fluorescent quantitative Real time-PCR method to determine of myocardial cell specificity of a-MHC genes mRNA expression level.Results1.In continuous culture first, third and fifth day, compared with nil group, normal serum containing cell activity showed no significant difference(P>0.05). Compared with nil group, only sf group ESC proliferative activity was significantly inhibited(P<0.05), the first, third and fifth day inhibition rate was 16%,8% and 6% respectively.2.At first day, compared with fbs group, nil group was no significant difference(P>0.05). Compared with nil group, st, sf, gt, gf group could promote ESC late apoptosis, the difference was statistically significant(P<0.05). At third and fifth day, compared with fbs group, nil group was no significant difference(P>0.05); but compared with nil group, st, sf group could promote late apoptosis, the difference was statistically significant(P<0.05).3.Compared with fbs group, nil group a-MHC mRNA expression was significantly lower(P<0.05); but compared with nil group, st, gt, gf group of a-MHC expression increased, sf group lower, the difference was statistically significant(P<0.05).Conclusion1.In this experiment dose [pinellia powder 3.45g/(kg·d)], Pinellia powder showed a significantly embryo toxicity, while Ginger ginseng pinellia decoction or powder had no obvious embryo toxicity.2.That Pinellia decoction or powder containing serum could promote ESC late apoptotic was a concrete manifestation of its inherent toxicity. The compatibility of Ginger ginseng pinellia decoction containing serum could promote the proliferation and differentiation of mouse ESC, presumably by "Ginger can restrict Pinellia toxicity" mechanism to achieve. That Ginger ginseng pinellia decoction or powder containing serum could raise a-MHC mRNA expression levels of differentiation stage mouse ESC in vitro experiment proves that Ginger ginseng pinellia or powder can be used to treat pregnancy vomiting science and rationality.3.As pregnancy contraindication medicine in the history of child health care of traditional Chinese medicine, Pinellia should be used with caution, but with pregnancy contraindication medicine should be used following the principle of "There is therefore not perish". |
