Objective The diagnosis of KOA based on clinical symptoms and imaging, these methods can not be diagnosis early and alwayscause serious consequences. This subject is to observe patients with knee osteoarthritis(KOA) in the urine of CTX- Ⅱ and pathological changes of Zn2 +, Ca2 + concentration and its degree of relationship. This study sought a conducive to the timely intervention and disease is simple, economic and effective diagnostic methods, get rid of the large imaging equipment and routine blood, more suitable for large-scale census, diagnosis of knee joint osteoarthritis.Methods Eighty two patients with KOA were treat as KOA group. At the same time we choose twenty healthy volunteers as control, that is group I.Urine sample and X rays of knee joint were collected.The images were classified according to Kellgren-Lawrence radiographic grading criterion. The patients were divided into group grade 0,group grade Ⅰ,group grade Ⅱ, grade III and grade IV. We use the Enzyme-linked immunosorbent assay(ELISA) to detect the concentration of CTX-Ⅱ in the urine.The concentration of Zn2+、Ca2+ in urine were detected by Inductively coupled plasma atomic emission spectrometry(ICP-AES). Result The concentration of CTX-Ⅱin group 0 was obviously lower than that in group IIV. Comparison the concentration of CTX-Ⅱ in each group :group IV[(372.590±16.865)pg/ml]>group III [(319.149±22.925)pg/ml]>groupⅡ [(243.065±26.069)pg/ml]>groupⅠ[(174.124±23.340pg/ml]andgroup 0 [(163.976±16.459)pg/ml],(F=334.402,P=0.000)(The concentration of CTX-Ⅱ in group Ⅰand control group 0 was no difference(p>0.05).The concentration of Zn2+ in urine in KOA group [(0.7000±0.1736) mg/L]was higher than that in control group[(0.3764±0.2163)mg/L],(F=53.401,P <0.000).But there was no difference in each KOA group(P>0.05). The concentration of Ca2+ in urine in KOA group [(204.3536±71.6828) mg/L]was higher than that in control group[(80.4635±39.3493)mg/L],(F=55.379,P <0.000).But there was no difference in each KOA group(P > 0.05). Conclusion The concentration of CTX-Ⅱ can indicate the progress of KOA. Little help for early diagnosis for KOA. The concentration of Zn2+、Ca2+ in urine is helpful for early diagnosis for KOA, but cannot indicate the progress of KOA. However,the concentration of CTX-Ⅱ combine with concentration of Zn2+、Ca2+ in urine may be helpful for early diagnosis for KOA.
