ObjectiveThe controlled clinical studies about osteoporosis (OP) and osteoarthritis (OA) in mitochondria. To explore the relationship between osteoarthritis and osteoporosis through variance analysis, and preliminary analysis of the possible mechanism.MethodsCollected 12 patients from March 2015 to December in Nanhai Hospital of traditional Chinese Medicine spine surgery, consistent with diagnostic crite ria, inclusion criteria, exclusion criteria of osteoporosis (OP), osteoarthri-tis(OA), osteoarthritis merger osteoporosis (OA&OP),and the control group (C). According to the order is divided into osteoporosis group (OP), osteo-arthritis (OA), osteoporosis with osteoarthritis (OA&OP) and control group (N).To comparing the difference between the groups of mitochondrial gene expression profile and preliminary analysis on genetic variations with the MeDIP-chip technology strategy and mass spectrum segmentation scans.To make a detailed record of all data and its statistical analysis.Results1. No statistical significant difference between groups of age. After correlation coefficient matrix analysis and DNA agarose gel Swimming analysis, specimens with strong consistency, and DNA fragment size conform to the requirements of the subsequent test.2. Comparison among groups genome methylation level:OA, OP, OA&OP exposure resulted in significant loss of DNA methylation compaired to control (P<0.05),and OA&OP exposure resulted in significant loss of DNA methylation compaired to OA, OP(P<0.05). But no significant difference between OA and OP(P>0.05).3. We filtered the enrichment from the OA and OP group separately, and focused on signals enriched in chronic group. Therefore,1222 regions were selected because they exhibited hypermethylatin in chronic exposed group and unmethylation in control and acute group. In order to identify functions associated with these differently methylated CpGs and genes, we performed GO and KEGG pathway analysis. The differential genome-wide promoter methylation regions involved in almost whole biological process, for example:protein metabolism, developmental process, regulation of gene expression:dorso-ventral axis formation, mTOR signaling pathway, NOD-like receptorsignaling pathway, etc.4. Comparison between groups in HCP and ICP methylation gene, compared with normal control group, OA group differences in gene has 644, OP group differences in gene has 669, OA&OP group differences in gene has 492. To meet with OA&OP group and normal control group at the same time compare the differences in gene analysis, found in the OA and OP group has four common genetic variations, specific for:PPIL3, NIF3L1, SMTN, CALHM2.GonclusionOsteoporosis and/or osteoarthritis genome methylation level is low. Common difference of osteoarthritis and osteoporosis reflects on some specific promoter, these genes involved in almost all biological processes, in order to further explore the theoretical basis for regulation of related effect. OA&OP group and normal control group with difference compare analysis and found that there are four common in OA and OP group differences in gene (PPIL3 NIF3L1 SMTN, CALHM2), may pass through the pathways involved in the disease process, provide clues for the correlation of the disease, is also likely to be passed to suppress the DNA methylation methods and drugs to provide important basis for the prevention and treatment.
