| ã€Objective】To investigate the biological function of mi R-4313 in the malignant transformation process of renal cell carcinoma, explore its mechanism, and provide new ideas and new methods of molecular targeted therapy in renal cell carcinoma.ã€Methods】Part I: 1. Using qRT-PCR to detect the expression of miR-324-3p and miR-4313 in renal cell carcinoma.2. MicroRNA mimics were transiently transfected into 786-O and OS-RC-2 cells. Then, we evaluated the cell proliferation using MTT assay in renal cell carcinoma cells. 3. MicroRNA mimics were transiently transfected to 786-O and OS-RC-2 cells. Then, we evaluated the cell migration and invasion using wound healing and transwell assay in renal cell carcinoma cells.Partâ…¡: 1. Predicted the potential gene targets of miRNA-4313 by gene target prediction softwares. 2. qRT-PCR technology was used to examine the expression of the candidate gene targets in 786-O cell transfected with increasing concentrations of miR-4313 mimic.ã€Results】Part I: 1.miR-324-3p and miR-4313 have a low expression levels in renal cell carcinoma. 2. The proliferation ability of renal cell carcinoma cells was weakened after they were transfected with miR-324-3p and miR-4313 mimic. However, miR-4313 has stronger ability to inhibit cell proliferation. 3. The migration and invasion ability of renal cell carcinoma cells were not weakened after they were transfected with miR-324-3p and miR-4313 mimic.Part II: 1. By gene target prediction softwares we found that UCP4, CUX1, MBD6 and TBX15 were candidate target genes of miR-4313. 2. qRT-PCR show that overexpression of the mi R-4313 mimic strongly decreased TBX15 mRNA expression. However, the remaining three candidate target gene mRNA levels did not change.ã€Conclusions】In the malignant transformation process of renal cell carcinoma, the expression of miR-4313 occurs to reduce, leading to weaken the negative regulation of target gene TBX15 and increasing the expression of target gene TBX15. The proliferation of renal cell carcinoma was promoted. Then renal cell carcinoma was obtained greater malignant phenotypes. Ultimately, renal cell carcinoma occurs to progression and malignant transformation. This experiment preliminary discussed the molecular mechanisms of progression of renal cell carcinoma. It aims to provide new ideas and new methods of molecular targeted therapy in cancer. |
