ObjectiveCentral obesity of menopause women is due to changes in the female of physiology, psychology and other aspects of abnormal distribution of body fat, which accumulates in the waist and abdomen mainly formed in the center of the waist and abdomen obese. Menopausal women central obesity has serious harm to the health of menopausal women. Studies have shown that:it is one of the important causes of menopausal women with type 2 diabetes. Chinese medicine believes that postmenopausal women with central obesity is the main pathogenesis of kidney yin and yang-based, supplemented by dampness. Therefore the choice Erxian Decoction of having a warm kidney yang, nourishing kidney yin and dispelling dampness. Western medicine believes that the main factors leading to the central obesity occurs in menopausal women is estrogen secretion decreased. So Western medicine choose estrogen replacement therapy for menopausal women to prevent and treat central obesity, but long-term use may increase the chances of post-menopausal women suffering from breast cancer and other diseases. Whether Chinese medicine Erxian Decoction choice can prevent menopausal women with central obesity and central obesity prevention menopausal women with type 2 diabetes caused it? In this study, based on the above questions:Investigating the effects of Erxian decoction on insulin sensitivity in ovariectomized rats and its mechanisms.MethodsFirst, on the overall level, to observe the effects of Erxian decoction on insulin sensitivity in ovariectomized rats.Fourty-female Sprague Dawley rats were overiectomized. On the seventh day, they were randomly divided into five groups of eight which were sham-operated group (SHAM), model group (OVX), low Erxian Decoction dose group (EXDL,30mg/kg), high Erxian Decoction dose group (EXDH,60mg/kg), estrogen group(E2,0. Olmg/kg). SHAM and OVX:normal saline, once a day. After 14 weeks of continuous treatment, we detected the relative content of each group of rats abdominal fat and subcutaneous fat by CT. Then, we tested the fasting blood glucose levels, the serum insulin levels, the glucose tolerance, and calculated the insulin resistance index. Finally, we detected the mRNA expression of MCP-1 and adiponectin in internal abdominal fat by qPCR.Second, at cellular level, to study the mechanism which Erxian decoction affects insulin sensitivity in ovariectomized ratsPreparated the serum containing Erxian Decoction. Fifteen female Sprague Dawley rats were randomly divided into three groups of five:namely control group (saline), EXDL(30mg/kg), EXDH(60mg/kg). To ecapitated blood, at room temperature for 2h,1300rpm,4℃, centrifuge 10min. Differentiated 3T3-L1 adipoctyes were cultured under anoxic box for0h,4h,12h,24h. We tested the MCP-1 levels in culture medium by ELISA. Then, three groups random divided: hypoxia control serum intervention (HCSI), hypoxia EXDL serum intervention(HEXDLSI), hypoxia EXDH serum intervention(HEXDHSI). Then, we applicated ELISA to detect MCP-1 content of each group.ResultsOn the overall level, Our results demonstrated that Erxian Decoction could reduce fat content of abdominal in the ovariotomy rats, insulin resistance index and improve glucose tolerance. qPCR detection showed that Erxian decoction could reduce MCP-1 mRNA level and increase Adiponectin mRNA level in intra-abdominal fat. However, at cellular lever,3T3-L1 fat cells induced by vitro hypoxic Oh,4 h,12 h,24 h could secrete MCP-1. After 3T3-L1 preadipoctyes differented were intervented by low and high dose of Erxian Decoction drug-containing serum for two hours, it was no statistical effection on the hypoxia 3T3-L1 preadipoctyes differented by comparing with control group.Cell level results:The 3T3-L1 preadipocytes induced differentiation and maturation in vitro hypoxia 0h、4h、12h、24h and ELISA test each time the cell supernatant MCP-1 concentrations results showed:The best time to hypoxia point They were:12h. ELISA detection of serum containing pre Erxian intervention 2h, hypoxia after 12h, cell supernatants in each group showed that MCP-1 levels:low and high dose Erxian Decoction containing serum pre-intervention after hypoxia-inducible 3T3-L1 fat cells secrete no significant difference compared to serum levels of MCP-1 pre-intervention and control group.ConclusionErxian Decoction could improve insulin sensitivity of ovariotomy rats, which may be related to its improvement the endocrine function of the intra-abdominal fat.3T3-L1 adipoctyes could secrete MCP-1 by Hypoxia inducion. Erxian Decoction-containing serum did not significantly reduce the MCP-1 level.
