| ObjectiveTo investigate the relation among the abnormal expression of Bub1 Protein and prostate cancer occurrence, development, metastasis and its clinical significance by observing the expression of Bub1 protein in prostate cancer and benign prostatic hyperplasia(BPH) tissues.Methods1 research objectsFrom May 2011 to May 2014, 118 cases of biopsy paraffin samples which has been confirmed as prostate cancer in The Affiliated Cancer Hospital of Zhengzhou University and 40 cases of benign prostatic hyperplasia(BPH) paraffin-embedded tissue samples operated in our hospital due to BPH at the same period were collected. The prostate cancer patients selected for the embedded tissue specimens were not receiving endocrine therapy, radiotherapy or chemotherapy before prostate biopsy. Among prostate cancer patients after diagnosis, 28 cases were performed radical prostatectomy(RP); 90 cases were treated with maximal androgen blockade(MAB), while patients with bone metastases were treated with bisphosphonates. Prostate cancer patients treated with radical prostatectomy were in close postoperative follow-up until the emergence of biochemical recurrence, local recurrence or distant metastasis; prostate cancer patients with maximal androgen blockade were followed up until disease aggravated to castration resistant prostate cancer.2 Experimental methodsThe expressions of Bub1 protein in prostate cancer and benign prostatic hyperplasia tissues were detected by immunohistochemical S-P method.3 Statistical analysisSpss20.0 statistical software were used for data statistical analysis. Measurement data(the age of two groups patients) was showed by X±S and comparison were tested with t. Comparison between different groups were used χ2 for test. P < 0.05 means the difference has statistical significance.Results1 Patients’ age in two groupsThe average age of patients with prostate cancer group and benign prostatic hyperplasia group were respectively(68.4±3.5)years old,(67.2±3.8)years old. There was no statistically significance among them.(P>0.05).2 Immunohistochemistry test results of Bub1 protein in patients of two groupsBub1 protein expression rate of prostate cancer and benign prostatic hyperplasia tissues were 48.3%(57/118), 77.5%(31/40), and there was significant difference between two groups(P <0.05).3 The relation between Bub1 protein expression and clinical features of prostate cancerBub1 protein expression of prostate cancer tissue was related to prostate cancer clinical staging, Gleason score and the time of prostate cancer progressed to castration-resistant prostate cancer. The later prostate cancer clinical staging, the lower rate of Bub1 protein positive expression in prostate cancer tissues will be. Compared with the positive expression rate of Bub1 protein in stage III and IV in prostate cancer, there was significant difference between them(P <0.05). The positive expression rate of Bub1 protein in prostate cancer tissues whose Gleason scores were less than 6 points, 7 points, and more than 8 points were 62.9%(22/35), 57.1%(24/42) 26.8%(11/41), and the difference was statistically significant(P <0.05).The positive expression rate of Bub1 protein decreased with the increase of Gleason score of prostate cancer; Among 90 cases of prostate cancer patients treated with the maximum androgen blockade, the positive expression rate of Bub1 protein who aggravated to castration resistant prostate cancer longer than and equal to 18 months or shorter than 18 months with prostate cancer were 66.7%(18/27) 39.7%(25/63), and the difference was statistically significant(P <0.05).Conclusions1 Compared with benign prostatic hyperplasia tissue the positive expression rate of Bub1 protein in prostate cancer tissue is obviously decrease.2 The later prostate cancer clinical staging, the lower rate of Bub1 protein positive expression will be.3 The higher Gleason score of prostate cancer, the lower rate of Bub1 protein positive expression will be.4 The shorter of the time prostate cancer progressed to castration-resistant prostate cancer, the lower rate of Bub1 protein positive expression will be. |
