| Objective: To investigate the pathological significance of HULC expression in hepatocelluar carcinoma and the relationship of epithelial-mesenchmal transition(EMT).Methods: All resected specimens of HCC were selected in Urumqi General Hospital of Lanzhou Military Region between April 2009-April 2015. SYBR Green real-time quantitative PCR detect the expression of HULC gene and correlate with the clinical pathological features,also analyze its relationship with Vimentin and E-cadherin protein.We determine the expression of Vimentin and E-cadherin protein in the 70 cases of HCC and adjacent liver tissues, 30 cases of surrounding liver tissue in hepatic hemangioma and liver trauma caused rupture by immunohistochemically analysed. Then analyze the relationships between the Vimentin,E-cadherin protein expression in HCC tissue and clinicopathological features of HCC patients.Results: Compared with the adjacent liver tissue, HULC gene expression increase in HCC(P=0.037), but vascular thrombosis was significantly higher than the adjacent liver group in the HCC group of HULC. Vimentin protein expression in HCC tissues was significantly higher than in adjacent liver tissues and benign liver tissue(P=0.000).E-cadherin protein expression in all tissues,but the expression was lower in HCC tissue than other tissue(P=0.018). Vimentin protein expression was negatively correlated to E-cadherin protein(r=-0.292,P=0.048).Futhermore, vascular thrombosis was significantly higher than the adjacent liver group in the HCC group of E-cadherin( P=0.008). HULC expression in HCC tissues was significantly higher in Vimentin expression tissue(P=0.029),and significantly higher in E-cadherin expression lower tissue(P=0.019).Conclusion: The high expression of HULC in HCCwas connected with the decreaseexpression of E-cadherin and the increase expression of Vimentin.It may prompting that HULCpromoted proliferation and invasion by EMT. |
