Objective:1.This study intends to determine elastin which express in connective tissue of vaginal anterior wall the Uygur and Han POP patients, to discuss the role of occurrence and development of pelvic support tissue structural and metabolic changes in POP. 2.To analysis and make clear whether the expression of elastin has difference between the two populations, analysis and summarizes the characteristics of the protein in gene expression and clinical significance. This study means a lot to clarify the POP etiology and racial diversity research,and plays a preventive role in etiology.Materials and Methods:1. Materials: Collected the pelvic organ prolapse patient who has treated in the First Affiliated Hospital of Xinjiang Medical University, gynecology 82 patients from July 2013 to December 2015 as Experimental group; 118 cases of hysterectomy due to benign lesions of the uterus were taken as control group. In the experimental group and control group, all the patients were married, had a history of vaginal delivery, corticosteroids and sex hormone drugs were not used at least three months before the operation. There were no significant differences in age, birth time, body mass index(P>0.05), has comparable. Diagnostic criteria is by the standard of POP-Q which was published by International Society for the control of urine. Specimens were collected by the hospital ethics committee for approval and access to the patients themselves or their families informed consent, and signed informed consent. Postoperative pathology experts confirmed the diagnosis.2. Methods:2.1 Using immunohistochemical method, the expression of elastin in the protein level of the anterior wall of the vagina was determined strictly according to the kit instructions. 2.2 Reverse transcription polymerase chain reaction(RT-PCR) method was used to detect the expression of elastin in the gene level strictly according to the kit instructions.Results:1. Elastin protein is expressed in the anterior wall of the vagina, The positive expression in the experimental group and the control group of Han respectively:(23.33)(86.66). The positive expression in the experimental group and the control group of Uygur respectively(16.66)(80), The expression of the experimental group was significantly less than that of the control group, the difference was statistically significant(p<0.05), the difference has no statistically significant(p>0.05).2. Agarose gel electrophoresis showed that: 143 bp for Elastin target band. The results of its semi-quantitative determination of the display that Elastin m RNA expression was positive in the four groups, Elastin m RNA displayed in the results semi-quantitative determination of electrophoretic that Han prolapse group expression level is( 0.402±0.145),Han control group expression level is(1.005±0.103),the Uygurs prolapse group expression level is( 0.390±0.094),the Uygurs control group expression level is(1.006±0.113),the difference was statistically significant compared in experimental group and control group(p<0.05); There was no significant difference between the two nations( p>0.05).Conclusion:1. Elastin protein express in all the anterior vaginal wall tissue, the difference in the experimental and control group has statistical significance, indicating that elastin protein in maintain supporting tissue of pelvic cavity functions play an important role, the in the occurrence of pelvic organ prolapse, development play an important role.2. Between the Uygur and Han in the experimental group and the Han group respectively difference were not statistically significant, indicating that the relevant factor in the pathogenesis of pelvic organ prolapse disease no racial differences, to provide support and assistance to the research on the etiology of pelvic floor dysfunction disease. |
