| ObjectiveTo investigate the elastin and elafin expression in cardinal ligament of women with Pelvie organ ProlaPse(POP) so as to determine the alteration that contributes to POP.Materials and methods1,Tissue samples and groupsFifty-one POP and seventeen NON-POP cardinal ligament and paries anterior vaginase tissue specimens were obtained from patients underwent surgical resection at Shengjing Hospital of China Medical University between 2007 October.and 2009 October. All the samples were fixed by formalin and embedded in paraffin.2,Reagentselafin mouse anti-human monoclonal antibody and elastin rabbit anti-human polyclonal antibodies were purchased from Santa U.S. companies, Diaminobenzidine (DAB) chromogenic agent SP kit were purchased from Beijing Zhong Shan Biotechnology Co, Ltd.3,MethodsImmunohistochemical staining was performed by the streptavidin-peroxidase (S-P) method. The sections were deparaffinized, water processed, treated by high tempreture to expose antigen and incubated with primary antibody at 4.0℃overnight. The color was developed with diaminobenzidine and the section were couterstained with harmatoxylin. For the nonspecific reagent control, the primary antibodies were replaced by PBS. Results found:extracellular matrix in see positive expression of brown-yellow granules. Light microscope slices in each of 10 randomly selected high power field, scoring criteria:0, no positive region; a positive regional area of 1-20%; 2, the positive area has 20-50%; 3, the positive size of the area " 50%.0-1:In order to elastin or elafin negative expression; 2-3:elastin or elafin expression.4,Statistical analysisApplication SPSS13.0 statistical analysis software, elastin or elafin protein expression and clinicopathological features of the relationship between POP using chi-square test, the relationship between elastin and elafin protein expression using Spearman correlation analysis; P<0.05 as statistically significant.Results1,elafin protein expression in the extracellular matrix, in the POP group of patients with anterior vaginal and uterine ligaments organization, elafin the main level of positive expression were 67%,63%.In the control group, expression levels were 30%,20%. There was a significant difference between the two (P=0.005,0.001). Statistical analysis showed that those associated with stress urinary incontinence (P=0.003,0.001) and the degree of prolapse heavier person (ⅡvsⅢ,Ⅳ,P=0.028,0.014),the main ligaments and vaginal uterine tissue elafin positive expression rate was significantly higher. The elafin protein expression and patient menopausal status (P=0.212,0.353) and the number of vaginal deliveries (P=1.000,0.164) has nothing to do.2,elastin is also expressed in the extracellular matrix, elastin in the POP group of the vaginal wall, the main ligament in expression levels were significantly lower than the control group (P=0.004,0.001), its expression levels in patients with POP-Q is closely related to indexing (ⅡvsⅢ,Ⅳ,P=0.005,0.012), while the patient is accompanied by stress urinary incontinence P=0.489,0.123), the number of vaginal deliveries (P=0.301,0.149) and in patients with menopausal status (P=0.841,0.696) has nothing to do.3,POP group of patients with anterior vaginal elafin and elastin for a total of 12 patients with positive expression, elafin and elastin are negative for 10 cases. The Spearman correlation analysis, elafin and elastin expression was not significantly correlated (P= 0.689), the main ligament of the organization in the womb there is no statistically significant (P= 0.105).Conclusions1. In POP group, the expression of elastin may be relative with the genesis and development of POP.2. elafin expression may be one of the causes leading to POP. |
