Objective:To investigate the correlation between plasma homocysteine (Hcy), Methionine synthase(MS)A2756G,Methionine synthase reductase(MSR)C524T polymorphisms and Essential Hypertension (EH) in Uygur, Han and Kazak ethnic in Xinjiang. Methods:433 cases including 179Uyghur,187Han and 67Kazak patients diagnosed by EH were collected by the First Affiliated Hospital of Xinjiang Medical University from September 2013 to July 2015, while 457 populations including 182Uyghur ethnic,196 Han ethnic and 79Kazak ethnic were studies as control. Polymerase Chain Reaction and Restriction Fragment Length Polymorphism (PCR-RELP) was adopted to detected the gene polymorphisms of MSA2756G, MSRC524T. Enzyme Immuno Assay was applied to measure the levels of plasma Hcy.chemiluminescence was applied to determine the plasma folic acid and Vitamin B12. Analysis the correlation of the biochemical indexes and MSA2756G, MSRC524T gene polymorphism with EH.Results:(1) Compared with the control, plasma Hcy levels were significiantly higher in EH group in both Uyghur, Han and Kazak ethnic(P<0.05). (2)There were no significant differences in MSA2756G single nucleotide polymorphism (SNP) between EH patients and controls in Uyghur,Han and Kazak ethnic(P>0.05).However,there were significant differences in MSR C524T single nucleotide polymorphism (SNP) between EH patients and controls in both Uyghur Han and Kazak ethnic(Uyghur:X2=6.513, P=0.03; Han:X2ï¼12.674,Pï¼0.002; Kazak:X2=11.358,Pï¼0,002). (3) There were no significant differences in plama homocysteine level carrying with AA〠AGã€GG of MSA2756G genetic locus between EH patients and controls in Uyghur,Han and Kazak ethnic(.P>0.05).However, plama homocysteine level carrying with MSR524C was statistically higher than that with MSR524T, (P<0.05). (4) After adjusting confounding factors such as gender, age, the logistic regression analysis indicated age (OR= 1.844 95%CI1.310-3.196, P=0.004),obesity (OR=2.691 95%CI 1.590-3.555, P=0.000), hyperhomocysteine (OR=1.53095%CI1.053-2.589, P=0.037) were independent risk factors for Uygur patients with EH, while age (OR=1.09795%CI1.021-1.706,P=0.018), hyperhomocysteine (OR=4.023 95%CI2,454-6.503, Pï¼0.000), obesity (OR=1.798 95%CI 1.127-3.361,p=0.007) were independent risk factors for EH patients in Han ethic. And age (OR=1.07395%CI0.989-1.207, P=0.032),obesity (OR=1.31495%CI 1.124-1.513,P=0.001), hyperhomocysteine(OR=2.264 95%CI 1.091-4.715,P=0.029) were independent risk factors for Kazak patients with EH.No correlation was found existed between the gene polymorphism of MS A2756QMSR C524T and EH patients of Uyghur,Han and Kazak nationality. Conclusion:Age, Obesity and hyperhomocysteine are independent risk factors for Uyghur,Han and Kazak ethnic with EH in Xinjiang respectively. The MSR 524T genotype can reduce the plasma concentration of Hey. It was no found that MSA2756G,MSRC524T gene polymorphisms were independent risk factors for Uyghur,Han and Kazak nationality with EH patients in Xinjiang. |
