Objective: Our study based on the inflammatory theory of aging,we plan to observe the effects on the middle-aged rats’ myocardial tissue inflammation by interposing aerobic exercise and curcumin.To explore the different interposed ways of the aging process myocardial proinflammatory cytokines in the elderly, we try to explain the effects of aerobic exercise combined with medication supplements myocardium in aging anti-inflammatory, in order to provide the new ideas that decreased the productions of inflammatory reaction among aging process.Methods: 40 rats,18-month-old, were used as experimental subjects, they were randomly divided into control group(QC, n = 10), curcumin group(CC, n = 10), the aerobic exercise group(OC, n = 10) and curcumin combined with aerobic exercise group(EC, n = 10). Group QC were perfused of 1% sodium carboxymethyl cellulose, they had not any exercise load, to maintain normal living conditions. Group CC perfusion corresponding concentrations of curcumin by weight, not applied sports. Group OC and EC group at the beginning of every day half past four p.m. no weight training treadmill at a speed of 15 m / min, 50 min / d, 5 days a week for training, to maintain this exercise program for 8 weeks. Group EC did not exercise until reperfused curcumin. After the training program was completed, the rats in the last 24 h weighed after exercise, anesthesia were killed, serum and heart tissue. Conventional staining Heart structure, ELISA(enzyme-linked immunosorbent assay) assay in serum and myocardial tumor necrosis factor-α(TNF-α), interleukin--1β(IL-1 β) concentration, Western blot(Western blot) measuring expression of myocardial NF-κBp65(nuclear factor-κBp65).Results:(1)The body weight of rats in each group were significantly decreased after the experiment. compared with the QC group, another groups’ weight and heart coefficient aerobic exercise group rats were significantly lower;(2) TNF-α:After eight weeks of the intervention, compared with QC,group CC rats serum concentrations of TNF-α a downward trend(P> 0.05),myocardial tissue concentrations of TNF-α in a downward trend(P> 0.05).Group OC serum concentration of TNF-α were significantly lower than in group QC(P<0.05), myocardial slightly lower levels of TNF-α(P>0.05).Group EC was significantly lower than that in group QC(P<0.05);Compared with CC,serum and myocardial tissue of rats in group OC of the concentration of TNF-α is slightly lower(P>0.05),EC group was significantly decreased P<0.05);compared with group OC,EC group was significantly increased(P<0.05);(3)IL-1β:After eight weeks of the intervention,compared with group QC,serum and myocardial tissue of rats in group CC IL-1β levels were significantly lower(P<0.05),EC group with a very significant difference(P<0.01),group C slightly lower(P>0.05);contrast with group CC,group OC serum and myocardial tissue IL-1β levels higher(P>0.05),EC group was significantly higher(P<0.05);compared with group OC,ECgroup significantly increased(P<0.05);(4)Compared with group QC, CC, myocardial tissue of rats in group OC NF- κ Bp65 concentrations were significantly decreased(P <0.05), EC group has a very significant difference(P <0.01).Compared with group CC, group OC myocardial tissue concentration of NF-κBp65 slightly higher, was not statistically significant(P> 0.05).Group EC was significantly decreased(P <0.05);compared with OC,ECgroup was significantly lower(P <0.05);Conclusions:(1)TNF-α, IL-1β content increase with age, so that the body en-ters a state of inflammatory aging.(2)Long-term aerobic exercise can reduce aging rats’ s weight, it’s TNF-α and IL-1β content was reduced, thereby reducing the body’s in-flammatory symptoms of aging.(3) Curcumin can inhibit NF-κB’s activity, decreased TNF-αand IL-1β’s level, caused by low expression of NF-κBp65 subunits,thereby inhibiting downstream gene transcription, reduce proinflammatory cytokines IL-1β and TNF-α’s expression.(4) Curcumin combined aerobic exercise can have better effects on reducing myocardial inflammation factor expressi on and thus play a more effective role in myocardial protec tion, delay anti-inflammatory aging. Those Tips us that pha rmaceutical supplements joint aerobic exercise can be used as a new way of life of aging.
