Objective:According to the "Expel the evils in muscles by means of diaphoresis" function indications and the anti-allergic effects of exterior-releasing herbs Ramulus Cinnamomi, the different extractions of Ramulus Cinnamomi were prepared with organic reagent extraction. To study the active extraction of Ramulus Cinnamomi, in vivo and in vitro research methods were used to conduct this experiment, and its anti-allergic mechanism was explored via Th1/Th2 imbalance, mast cell degranulation, and IgE release, This work provides the modem pharmacoiogical basis for the treatment of allergic disease with "divergent wind chill" medicine.Methods:① Delayed type hypersensitivity mouse model induced by DNCB and heterogeneous passive cutaneous ana-phylaxis mouse model were used to evaluate the effects on capillary permeability and swelling of the ear of the different extractions from Ramulus Cinnamomi. And the hyaluronidase inhibition binding experiments in vitro also had been done to preliminary screening Ramulus Cinnamomi anti-allergic extraction. ② then the anti-allergy effect of decoction without essential oil and ethyl acetate extraction from Ramuius Cinnamomi was clarified by histamine and serotonin-induced increasing of skin capillary permeability mouse model and ovalbumin-induced asthmatic guinea pig model. ③ the content of IFN-γ. IL-4, and IgE in serum, and IFN-γ, IL-4, IL-18, total protein levels in alveolar lavage fluid of bronchial asthma model guinea pig were determined. And the β-hexosaminidase and histamine release rate of rat peritoneal mast cells affected by containing serum was observed in vitro. Aim of the above studies was tc study the anti-allergic mechanism of effective extraction of Ramulus Cinnamomi. ④ at last, the categories of active extraction from Ramulus Cinnamomi were preliminary researched by physical and chemical reactionsResults:① the animals were administered with the decoction without essential oil (6g/kg), ethyl acetate extraction (6g/kg), and water extraction (6g/kg) of Ramulus Cinnamomi for 10d can significantly reduce the capillary permeability (P<0.01 or P<0.05) of delayed type hypersensitivity mice. And the ethyl acetate fraction of Ramulus Cinnamomi also can inhibit the ear swelling of the delayed type hypersensitivity mice (P<0.05); Vascular permeability was significantly inhibited (P<0.05 or P<0.01) of heterologous passive cutaneous anaphylaxis in mice for orally administered 7d. The decoction without essential oil and ethyl acetate extraction from Ramulus Cinnamomi had a strong inhibition of hyaluronidase, the inhibition rate was 52.48%. 52.17% respectively.② The decoction without essential oil (6g/kg) and ethyl acetate extraction (6g/kg) from Ramulus Cinnamomi can significantly inhibited (P<0.01, P<0.05) capillary permeability increase of histamine and serotonin-induced increasing of skin capillary permeability mouse; Administered orally for 7d. Ethyl acetate extraction (1.665 and 3.33g/kg) and decoction without essential oil (1.665g/kg) from Ramulus Cinnamomi had a significant reduction of white blood cell count of bronchial asthma model guinea pig, reducing guinea pig lung tissue eosinophils infiltration, and having a tread to relieve of cell debris, epithelial cell loss, and inflammatory cells in the bronchial lumen of lung tissue, the total protein content in bronchoalveolar lavage fluid can also be reduced. ③ after administered orally ethyl acetate extraction (1.665 and 3.33g/kg) and decoction without essential oil (1.665g/kg) from Ramulus Cinnamomi for 7d, the level of IFN-γ was increased and the content of IL-4 was reduced in bronchoalveolar iavage fluid and serum from bronchial asthma model guinea pig with varying degrees. And the content of IL-18 in lavage fluid was increased (P<0.01), while IgE content of serum was reduced (P<0.01). The containing serum of ethyl acetate fraction and decoction without essential oil can inhibit the releasing of β-hexosaminidase and histamine from rat peritoneal mast cells, having inhibited mast cell off degranulation active in vitro.Conclusion:Decoction without essential oil and ethyl acetate extraction from Ramulus Cinnamomi were the effective extraction parts with anti-allergic active. Water soluble component in Ramulus Cinnamomi played a dominant role in the anti-allergic effect. Ramulus Cinnamomi extractions anti-allergic effect was achieved through multiple channels, including corrected imbalance of Th1/Th2 cell subsets by inhibition of cytokine IL-4 release and promotion of IFN-γ, IL-18 cytokine release, inhibition of mast cell degranulation process and reduce the release of allergy mediators, inhibiting IgE generation, and reducing the infiltration of eosinophils etc.
