| Sini Decoction(SND) is a famous formula from<Shang Han Za Bing Lun> which is composed by Aconiti Lateralis Radix Praeparata (Fuzi), Zingiberis Rhizoma (Ganjiang), Glycyrrhizae Radix et Rhizoma (Gancao) and praised as the first formula of reviving yang for resuscitation. Now it is also recorded by the current version of <Chinese Pharmacopoeia>. Aiming at the problem of attaching importance to herb-pairs and neglecting formula in the current research of SND, in this paper, we research the change rules of effective components and the biological effects expression in Acute Myocardial Infarction (AMI) rat of SND before and after compatibilities to overall reveal the influence of the combination of Fuzi-Ganjiang-Gancao on SND in aspects of chemical constituent and pharmacological effect, and to illustrate the scientificity of the medicine-using of SND.In the study of the change rules of the effective components of SND before and after compatibilities, we take SND, different medicine-pairs decoction and different single medicine decoction as test samples and use HPLC to measure the contents change of effective components. The results show that the contents of four effective components of Gancao (liquiritin, glycyrrhizic acid, liquiritigenin, isoliquiritigenin) decrease after compatibilities and the sequence is Gancao Decoction> Gancao-Ganjing Decoction>Gancao-Fuzi Decoction>SND; the contents of three gingerol components of Ganjiang (6-gingerol,8-gingerol,6-shogaol) increase after compatibilities and the sequence is Ganjiang Decoction> SND> Ganjiang-Fuzi Decoction> Ganjiang Decoction; the contents of three mono-ester aconitum alkaloids of Fuzi (benzoylaconitine, benzoylhypacoitine, benzoylmesaconine) increase after compatibilities and the sequence is SND>Fuzi-Gancao Decoction>Fuzi-Ganjiang Decoction> Fuzi Decoction. These results illustrate that the effective components of SND change following some rules in the compatibility process and further reveal the mechanism of mutual-restraint/mutual-detoxication, mutual-promotion/ mutual-assistance compatibilities of SND from the chemical composition angle.In the study of the pharmacodynamic differences of SND before and after compatibilities, we establish the AMI rat model and research the influence of SND, different medicine-pairs decoction and different single medicine decoction on AMI rat myocardial infarction size and related biochemical indexes. The results show that all of the decoctions can decrease the myocardial infarction size and restrain the abnormal expression of SOD, GSH-Px, MDA, LDH, CK, AST of AMI rat in a certain degree. Compared with the model group, the Ganjiang group, Gancao group, Ganjiang-Gancao group do not show significant difference while the SND group, Fuzi-Ganjiang group, Fuzi-Gancao group and Fuzi group show significant difference (P<0.05 or P<0.01). The sequence of pharmacodynamic effect strength is SND> Fuzi-Ganjiang Decoction>Fuzi-Gancao Decoction>Fuzi Decoction, which illustrate that Fuzi is the main medicine of the anti-myocardial ischemia effect of SND and the combination of Fuzi-Ganjiang-Gancao avail the efficacy expression of SND.The innovation point of this paper is that we discuss the change of effective components and biological effects expression of SND before and after compatibilities systematic, and reveal the scientific connotation of SND compatibilities from the medicine-pairs study angle and the whole formula study angle. By the comprehensive analysis of study results, we further put forward the reducing toxicity and enhancing efficacy mechanism in vitro of SND is that the interaction between Fuzi, Ganjiang, Gancao contribute to the dissolving out of alkaloids and gingerol, and the dissolved alkaloids switch between the combined state and free state constantly, which increase the content of the mono-ester aconitum alkaloids and decrease the content diester-alkaloids to achieve the goal of reducing toxicity and enhancing efficacy of SND compatibilities. |
