Objectives:To study the association between the three miRNA single nucleotide polymorphisms and prognosis of gastrointestinal cancer.Methods:We conducted a meta-analysis which concerning the association of microRNA(miR-608、miR-219-1、miR-423) polymorphisms with the prognosis of gastrointestinal tumor in Cochrane Library, PubMed, Embase, Web of Science, SinoMed and the CNKI database. We used Stata/MP software 13.1 to analyze the data from each study. Pooled Hazard Ratios with 95%Confidence Intervals for the relationship between three genetic polymorphisms and Overall Survival (OS), Recurrence-free Survival (RFS) were calculated. Subgroup analyses by population, type of tumor were conducted.Results:Stage Ⅲ colorectal cancer patients carrying the variant G allele of rs4919510 and receiving 5-FU chemotherapy were associated with a significantly worse survival. No significant results were obtained for miR-608 polymorphism associated with recurrence-free survival of colorectal cancer. T-containing genotypes of miR-219-1 act as a risk role for overall survival of colorectal cancer. The association between miR-423 polymorphism and overall survival was observed in Chinese population in CA vs. CC model.Conclusions:G-containing genotypes of mir-608 and T-containing genotypes of miR-219-1 act as a risk role for overall survival in patients with stage III CRC treated with fluoropyrymidine-based chemotherapy. CA genotype of miR-423 may be the risk factor for overall survival in Chinese population with esophageal squamous cell carcinoma and colorectal cancer.
