Objective: To observe the effect of Buyang Huanwu decoction on angiogenesis and after focal cerebral ischemia in rats,and to explore the underlying mechanism.Method: The rat middle cerebral artery occlusion models with filament of focal cerebral ischemia/reperfusion were established. The adult SD male rats were divided randomly into sham group, cerebral ischemiareperfusion model group and the Buyang Huanwu decoction group. The Buyang Huanwu decoction group was further divided into two sub-groups which were dealt with the low and high dose of Buyang Huanwu decoction. The neurological scores, cerebral infarction volume and the microvessel density were detected after ischemia. Moreover, the expression of Ang-1 m RNA were evaluated by RT-PCR,and the level of Ang-1 protein were evaluated by western-blot.Results: Compared with the model group,the neurological scores and brain infarct volume were decreased.The microvessel density were increased in the Buyang Huanwu decoction groups. In addition, the expression of Ang-1 m RNA and Ang-1 protein were significantly increased in two doses of Buyang Huanwu decoction groups.Conclusion: 1. Buyang Huanwu decoction can reduce brain infarction volume, have the effect that reduce brain damage. 2. Buyang Huanwu decoction can reduce the neurological deficit. 3. Compared with the sham-operated group and cerebral ischemia-reperfusion model group, Buyang Huanwu decoction can increase the microvessel density.4. Buyang Huanwu decoction can upregulation the expression of Ang-1 m RNA, and the expression of Ang-1 protein are in agreement with the results of the variation of Ang-1 m RNA. In conclusion,the results of the present study show that Buyang Huanwu decoction modulates angiogenesis and increases the expressions of Ang-1,may be one of the mechanism by which Buyang Huanwu decoction protect the ischemia reperfusion injury in rats. |
