Projects:Fracture repair process is regulated by many factors and regeneration of vascular networks is crucial. Intermittent injections of small doses of PTH can promote bone anabolic effect and promote fracture healing. However, relationship between PTH and angiogenesis is still unclear. VEGF that is an important angiogenic factor plays an important regulatory role in the process of angiogenesis. To explore whether the lack of endogenous PTH by reducing the expression of VEGF causes angiogenesis weakened during fracture healing and delays fracture healing.Methods:We constructed PTH knockouted mice fracture model and observe fracture healing from X-ray film and use immunohistochemistry to detect bone anabolic way and angiogenesis index.Results:PTHKO declines in VEGF expression in mice. PEGAM immunohistochemical positive reduces. OCN activity decreases and Fracture healing delays.Conclusion:we believe that lack of endogenous PTH may release VEGF by reducing mesenchymal stem cells or osteoblasts, reduce vascular fracture, inhibite osteoblastic activity and affect endochondral bone leading to delayed fracture healing eventually. |
