| Objective:To determine the main role of growth factor in different periods of fracture healing process of Yougui Yin and brain injury.To clear and definite physiological mechanism relationship between heart,small intestine and bone,and then obtain the modern analytic theory of TCM viscera.To obtain the best dosage of Yougui Yin to prevention fracturesMethods:1.Establish the rat models of traumatic brain injury and tibia fractures.The 186 rats are randomly divided into 7 groups,30 rats in each group:control group,brain injury group,fracture group,brain injury plus fracture group,low dosage of Yougui Yin plus fracture group,moderate dosage of Yougui Yin plus fracture group,high dosage of Yougui Yin plus fracture group.3 days after the operation of the control group,take out the callus and duodenal of the other 6 groups at the 3rd day,7th day,14th day,21st day,28th day.2.Screening out genes with significant differences by gene chip technology.3.Validate the changes of gene expression through the q-PCR,Western Blot and immunohistochemical stainingResults:1.The gene chip results showed that compared the model group with the control group,the PDGF、CGRP、VEGF、BFGF expressions of callus and the duodenum were significantly increase(R>2),which had a statistical significant.According to the results of Q-PCR,Western Blot and immunohistochemical staining,the trend of each index expressions of callus and the duodenum are the same.In callus and duodenal in fracture group,duodenal in brain injury group,the expression of PDGF rules is that begin to express at the 3rd day,gradually increase at the 7th day,reach the peak at the 14th day,then decline;The expression of CGRP rules is that begin to express at the 3rd day,gradually increase at the 7th day,reach the peak at the 14th day,then decline;The expression of BFGF rules is that express obviously at the 3rd day,peak at the 7th day,then decreas gradually;The expression of VEGF rules is that begin to express at the 3rd day,gradually increase at the 7th day,continue to strengthen at the 14th day,reach the peak at the 21st day,decrease gradually at the 28th day.PDGF,CGRP,VEGF,BFGF expression in Brain injury plus fracture group and Yougui Yin plus fracture group at callus and duodenum is significantly increased(p<0.05),significantly advance of the peak time(p<0.05),and maintained over time peak than brain injury group and fracture group.The indexes in Brain injury plus fracture group combined with high dosage Yougui Yin plus fracture group had no significant difference(p>0.05),but there is statistical significant differences among high,moderate and small dosage(p<0.05).The expression of PDGF rules is that begin to express at the 3rd day,get close to the peak at the 7th day,reach the peak at the 14th day,obviously decrease at the 21st day,get to the simple fracture group level at the 28th day;The expression of CGRP rules is that begin to express at the 3rd days get close to the peak at the 7th day,reach the peak at the 14th day,obviously decrease at the 21st day,get to the simple fracture group level at the 28th day;The expression of BFGF rules is that obviously express at the 3rd day,reach the peak at the 7th day,decrease slightly at the 14th day,decrease significantly at the 21st day,get to the simple fracture group level at the 28th day;The expression of VEGF rules is that begin to express,then increase gradually,reach the peak at the 21st day,then decrease.Conclusion:1.The high expression of PDGF、BFGF、CGRP、VEGF in brain injury plus fracture group and Yougui Yin plus fracture group proves that PDGF、BFGF、CGRP、VEGF are the main growth factors to accelerate callus formation.2.The growth factors of PDGF、BFGF、CGRP、VEGF play a major role in the early fracture healing process.The growth factors of PDGF、CGRP、VEGF play a major role in the mid fracture healing process.The growth factors of VEGF play a major role in the late fracture healing process.3.Heart(brain)-small intestine-bone is contact by PDGF、BFGF、CGRP、VEGF.4.In Yougui Yin group,the expression of PDGF、BFGF、CGRP、VEGF increased with the dosage increasing.It shows that high dosage is the best dosage to treatment. |
