Objective:To explore serum complement component 5(C5)levels and function in Uygur patients with pigeon fancier’s lung(PFL),and to evaluate the association of complement C5 gene polymorphism with susceptibility to PFL in Uygur people.Mehthod:92 cases were enrolled and serum samples were collected according to the criteria for inclusion and exclusion( 32 Uygur patients with diagnosis of Pigeon fancier’s lung(PFL) were included in Xinjiang Uygur region people hospital from January,2012 to December,2013,the negative control group was composed by 30 healthy bird-exposed but asymptomatic subjects, and the normal controls included 30 healthy people ethnically, without history of bird-exposed).Serum C5 levels were tested by enzyme-linked immunosorbent assay(ELISA).The C5 content changes were evaluated.Genomic DNA was extracted from the rest peripheral blood,we genotyped 14 single nucleotide polymorphisms(SNPs) in C5 gene polymorphisms by polymerase chain reaction(PCR)method, using Mass ARRAY Assay Design. The relationship of C5 gene polymorphisms and risk factors in Uygur patients with PBL was investigated by Chi-square test.Results:Serum levels of C5 were significantly higher in case subjects than in normal control subjects(P<0.04), C5 serum levels were also higher in case subjects than in negative control subjects(P<0.04).According to Hardy-Weinberning balancing test.Three single nucleotide polymorphisms( SNPs)in the C5 gene were found in our study : rs17611、rs17612、rs17216529.Alleles A、G and Genotypes GG、GA、AA were found in rs17611 and rs17216529; alleles C、A and genotypes CC、AA、CA were found in rs17612;allele frequencies A、G and genotypes GG、GA、AA in rs17611 showed no significant differences among three groups(χ2=1.18,P=0.554,χ2=4.445,P=0.349),it demonstrated C5 rs 17611 was not associated with the risk of PFL; allele frequencies A、C and genotypes CC、CA、AA in rs17612 showed no significant differences among three groups(χ2=3.056,P=0.217,χ2=3.144,P=0.218),it demonstrated C5 rs 17612 was not associated with the risk of PFL; allele frequencies A 、 G and genotypes GG、GA、AA in rs17216529 showed no significant differences among three groups(χ2=0.064,P=0.968,χ2=0.079,P=0.961),it demonstrated C5 rs 17216529 was not associated with the risk of PFL.Conclusions: Our study tested serum complement C5 levels and detected C5 gene SNPs to explore the association with PFL in Uygur people.We found the serum levels of C5 were much higher in case group than in the negative control and normal control groups,it indicated complement C5 may regulate immune reaction in the mechanism of PFL.14 sites were found in complement C5,Three SNPs in the C5 gene were found in our study: rs17611、rs17612、rs17216529.but all didn’t show any association with PFL,which may relate to C5 gene highly conserved.furthermore,small samples effected also.large population—based studies are still necessary to repeatedly confirm the findings.
