| Objective:To study the association between EPO Gene polymorphisms and type 2 diabetic retinopathy in Chinese Han population Fujian region. Meanwhile, analyzing the risk factors for type 2 DR,to offer a new way for preventing and curing type 2 DR.Methods: 256 cases of Type 2 diabetes mellitus who were diagnosed in the First affiliated Hospital of Fujian Medical University from September 2013 to August 2014 were collected, including 128 cases of diabetic DR and 128 cases of NDR.gender, age, height, weight, blood pressure, and diabetes duration were recorded. 8h Fasting peripheral venous blood were collected for 5ml and 2ml, 5ml of venous blood was used for the determination of clinical indicators, 2ml venous blood was used for DNA extraction, and then, the polymorphism of the two SNPs of EPO gene:rs551238 and rs1617640 were determined by Matrix assisted laser desorption ionization time of flight mass spectrometry. Analyze the information of DR group and NDR group with SPSS19.0 software, Analysis of the clinical index by using the method of analysis of independent samples T test and Logistic regression analysis, gene polymorphism with the chi square test(P <0.05).Results:1.clinical data: Age and duration of diabetes between the two groups had significant difference(P<0.05), Other clinical indicators was not statistically significant(P>0.05).Furthermore, the two groups were divided into ≤59, 60 ~69 and ≥70 three age groups, compared and analyzed the age groups of ≤59 and 60 ~69 years old with ≥70,respectively,the OR values were 8.667 and 5.667; Similarly, the two groups were divided into 1~9,10~19 and ≥20 three time periods, compared and analyzed the time periods of 10~19 and ≥20 with 1~9,respectively, the OR values were 5.556 and 14.44 times.2.Analysis of Hardy-Weinberg: The EPO gene rs1617640 and rs551238 loci by Hardy- Weinberg equation to test showed that all P values were greater than 0.05, indicating the difference was not statistically significant, its distribution accords with the Hardy-Weinberg balance, and has the group representative. 3.Comparative analysis of rs551238 polymorphism of EPO gene:the frequency of genotype in group NDR and DR were 0.02%ã€0.28%ã€0.70% and 0.09%ã€0.31%ã€0.59% for CCã€CA and AA respectively. the difference of the frequency was statistically significant(P=0.014, P < 0.05),and the OR value was 0.577.Furthermore,compared genotype of CA and AA with CC respectively showed that the difference was statistically significant, and the OR value were 0.18 and 0.141;Similarly, the difference of the allele frequency between two groups was statistically significant(P=0.014, P < 0.05),and the OR value 0.556. 4.Comparative analysis of rs1617640 polymorphism of EPO gene:the frequency of genotype in group NDR and DR were 71.87%ã€28.13%ã€0.00% and 60.15%ã€31.25%〠8.60% for TT〠TG and GG respectively. the difference of the frequency was statistically significant(P < 0.001),and the OR value was1.923. Similarly, the difference of the allele frequency between two groups was statistically significant(P=0.003 P < 0.05),and the OR value was 1.953. 5.Analyze the age, duration of diabetes mellitus, EPO gene rs551238 and rs1617640 genotype which were statistically significant by Single factor Logistic regression analysis with variable factor Logistic regression analysis showed that age, duration of diabetes mellitus, EPO gene rs1617640 genotype were statistically significant, and EPO gene rs551238 genotype was not.Conclusions: The age, duration of diabetes mellitus, EPO gene rs1617640 site TT/GT genotype and allele T all have correlation with type 2 DR. The younger age(between age40 ~ 91), the longer duration of diabetes mellitus(expecially more than 10 years),and carrying TT/GT genotype and T allele of the EPO gene rs1617640 site were related to higher risk of diabetic retinopathy. |
