| [Objective] To investigate the correlation between serum osteoprotegerin (OPG) level with its single nucleotide polymorphisms (SNP950) and type2diabetes (T2DM) with its retinopathy (NPDR)ã€Object】247cases and197cases randomly selected in Yanbian area were grouped into normal control group (NC,50cases), T2DM group (NDR108cases and BDR55cases), and PDR group (PDR34cases).ã€Methods】 Serum OPG levels were measured by enzyme-linked immunosorbent assay (ELISA); DRP was determined by ophthalmoscope examination; Taqman probe method for SNP genotyping, fluorescent sequencing method for the determination of gene sequences; and the related clinical indicators was observed and recorded. All data are presented as means±SD. The statistical analysis was performed using SPSS17.0.The comparison of the groups was done by variance analysis and pairwise comparison. The Correlation among variables was determined by using spearman related analysis; multivariate analysis was performed by using logistic regression analysis of risk factors for screening retinopathy. P-values<0.05were considered significantã€Results】1. Average serum OPG in the T2DM group was significantly higher than that in the NC group (2.16±0.40) pg/ml, while the PDR group (2.66±0.09pg/mL) was higher than that in the BDR group (2.43±0.05pg/mL) and NDR group (2.32±0.10pg/mL)(P<0.01). The BDR group was significantly higher than the NDR group (P<0.01). In the T2DM group the SBP, DBP, FPG2hPG and HbAlc index were significantly higher than that in the NC group (P<0.01). The rest of the indicators were not significantly different (P>0.05).2. The serum OPG of Diabetic retinopathy patients was significantly correlated with FPG,2hPG, HbAlc SBP and DBP (P<0.05), however not significantly correlated with BMI, TG, LDL-C HDL-C and CHO. 3. Line Logistic regression analysis was performed (diabetic group was treated as population; diabetic retinopathy as the dependent variable; age, disease duration, BMI, FPG, serum OPG levels and OPG genotype as independent variable). The results showed the occurrence of diabetic retinopathy was related with HbAlc,2hPG and serum OPG levels, and serum OPG levels was an independent risk factors for type2diabetic retinopathy4. According to the OPG gene, Patients with type2diabetes were divided into CC, TC and TT groups, the results showed that the serum OPG levels in the CC genotype group (2.50±0.32pg/mL) was significantly higher than the TT genotype (2.16±0.18pg/mL) and TC (2.28±0.28pg/mL) genotype group (P<0.05). The rest of the indicators were not significant different (P>0.05).5. TC type was the main genotype in the NC group and diabetic retinopathy group. CC genotype in the PDR+the BDR group (29.2%) was significantly higher than the NC group (12.0%)(P<0.05). C gene frequency in PDR the BDR group (54.5%) was significantly higher than that in the NC group (38.0%)(P<0.05).ã€Conclusions】1. Serum OPG levels are associated with T2DM and its severit y of retinopathy.2. OPG gene SNP950T-C is correlated with T2DM and its DRP, and C allele gene may be a risk factor for DRP.3. Serum OPG levels,2hPG, and HbAlc are risk factors for T2MD, and serum OPG level is an independent risk factor. |
