| Objective:To observe the intervention of atorvastatin on ulcerative colitis in mice and whether play a protective role of vascular endothelial by reducing the expression level of inflammatory cytokines.Methods:27 clean male C57BL/6 mice were randomly divided into 3 groups, the control group,model group and interventional group respectively has 15 rats.The control group drank sterile distilled water.The model group and interventional group freely consumed 2%DSS solution,and giving interventional group atorvastatin(10mg/kg·d) to fill the stomach in the week before modeling, at the same time giving model group equal volume of normal saline to fill the stomach.During the experiment, we have to compare each group the general condition and body weight change. After the end of the experiment, we observe the pathological changes in colon tissue of each group by HE staining, and measure serum levels of IL-8 and VEGF by ELISA method, and evaluate the levels of NF- ? B and VEGF by immunohistochemical method.Results:1.During the experiment, the control group were generally in good condition, the colon tissue structure was complete by HE staining. Compared with the control group, model group were in poor condition, intestine mucosa inflammation of model group was obvious and their serum levels of IL-8 and VEGF were significantly increased(P<0.05),The expression of NF-? B and VEGF of aorta were also increased by immunohistochemistry method.2.Compared with the model group, the general and colon histopathology performance of intervention group were significantly improved, serum levels of IL-8 and VEGF were significantly lower(P<0.05). The expression of NF- ? B and VEGF of aorta were also lower by immunohistochemistry method(P<0.05).Conclusions:Atorvastatin can reduce the expression of inflammatory cytokines in mice with ulcerative colitis to improve intestinal inflammation,and protect vascular endothelial by reducing the level of expression of inflammatory cytokines. |
