0bjective:By applying echocardiography combined with load test, the experiment was conducted to evaluate left ventricular diastolic function and the structure of left heart of the New Zealand white rabbit.Methods:The experiment was conducted to choose healthy male New Zealand white rabbits(n =20). Animals that are prepared of liver cirrhosis rabbit model by carbon tetrachloride(CCl4) composite pathogenic factor are divided into control group and liver cirrhosis group using self-contrast method. Appling GE-Vivid7 color ultrasonic diagnostic instrument and synchronous ecg gather M type echocardiography, mitral valve blood flow doppler spectrum diagram, tissue doppler image and the required indicators, the experiment was conducted to evaluate left ventricular diastolic function and analysis relations of left heart diastolic dysfunction to liver cirrhosis.Results:1. The comparison of heart rate: when injected dose was 5, 10 and 20 ug/(kg. min)respectively, compared with resting-state, there were no significant difference for two experimental groups heart rate increase(P>0.05); When injected dose reached 30 and 40ug/(kg. min) respectively, heart rate increased significantly(P<0.05). And heart rates of the cirrhosis group rabbit were higher than the control group under every dose of dobutamine(including resting-state).2. Left ventricular structure parameter changes: Four indicators(IVS, LVPWd,LVEDD and LVEDV respectively) of two groups increased with the increase of the dose.At the same dose(including resting-state), IVS, LVPWd, LVEDD and LVEDV of the cirrhosis group rabbit were significantly higher than the control group(P<0.05).3. Left ventricular diastolic function parameters: the control group rabbit E and Em were gradually rising with the increase of dose, which appeared increasing trend. E and Em rose gradually with small dose injection for the cirrhosis group rabbit; But when injected dose reached 30 ug/(kg. min), E and Em fell rapidly; When injected dose reached 40ug/(kg. min), E and Em significantly were lower(P<0.05) than resting-state and reached at a minimum value.When injected dose was 0, 5, 10 and 20 ug/(kg. min) respectively, there were no significant difference(P>0.05) for two groups E and Em; And when dobutamine injected dose reached 30 ug/(kg. min), the control group rabbit E and Em were significantly faster than the cirrhosis group(P<0.05). Two experimental groups rabbit A and Am gradually rose with the increase of dose. In addition, there were no significant difference under the same loading dose for two experimental groups rabbit A and Am(P>0.05).The control group rabbits E/A and Em/Am were greater than 1 under different dose(including resting-state), but its trend were irregular. The cirrhosis group rabbits E/A and Em/Am were greater than 1 under the resting-state and small dose injection(have not reached 30 ug/(kg. min)); But When injected dose reached 30 and 40 ug/(kg. min)respectively, E/A and Em/Am were significantly lower than the resting state(P<0.05), and were significantly lower than control group in the same load status value(P<0.05).Conclusions:1. The cirrhosis group rabbit left ventricular diastolic function weaken, especially in large dose injection.2. Compared with the control group, the cirrhosis group rabbit had poor tolerance to dobutamine.3. Dobutamine load test combined with echocardiography could improve detectable rate of the early cirrhosis left heart dysfunction. |
